Optimizing Prednisolone Loading into Distiller's Dried Grain Kafirin Microparticles, and In vitro Release for Oral Delivery

Kafirin microparticles have potential as colon-targeted delivery systems because of their ability to protect encapsulated material from digestive processes of the upper gastrointestinal tract (GIT). The aim was to optimize prednisolone loading into kafirin microparticles, and investigate their potential as an oral delivery system. Response surface methodology (RSM) was used to predict the optimal formulation of prednisolone loaded microparticles. Prednisolone release from the microparticles was measured in simulated conditions of the GIT. The RSM models were inadequate for predicting the relationship between starting quantities of kafirin and prednisolone, and prednisolone loading into microparticles. Compared to prednisolone released in the simulated gastric and small intestinal conditions, no additional drug release was observed in simulated colonic conditions. Hence, more insight into factors affecting drug loading into kafirin microparticles is required to improve the robustness of the RSM model. This present method of formulating prednisolone-loaded kafirin microparticles is unlikely to offer clinical benefits over commercially available dosage forms. Nevertheless, the overall amount of prednisolone released from the kafirin microparticles in conditions simulating the human GIT demonstrates their ability to prevent the release of entrapped core material. Further work developing the formulation methods may result in a delivery system that targets the lower GIT.

Summary: The concentration of dietary fibre plays a critical role in the composition and metabolism of gut microbiota. This study investigated the effects of different concentrations of fructo-oligosaccharides and pectin on microbial diversity, loads, and metabolite profiles. The results showed that high concentrations of dietary fibre increased the abundance of Methanobrevibacter genus and the production of butyrate. Understanding the underlying mechanisms allows for personalized modulation of gut microbiota by adjusting dietary fibre concentrations.

Phenolic and carotenoid characterization of the ethanol extract of an Australian native plant Haemodorum spicatum

Summary: This study characterized the phenolic and carotenoid profiles of the Australian native bulb plant, Haemodorum spicatum, using HPLC-DAD-ESI-QTOF-MS/MS and HPLC-DAD. Particle size of dietary fibre has diverse effects on in vitro gut fermentation rate and end-products depending on food source

Summary: This study investigated the relationships between particle size, composition, and fermentability of dietary fiber (DF) from various food sources. The results showed that different substrates had diverse fermentation outcomes regardless of particle size. Particle size differences did not consistently affect fermentation outcomes for different types of food. The nature of the particles and their major chemical components together determined the influences of particle size on fermentation.

Summary: Upon wetting, chia nutlets produce a gel-like capsule of mucilage, which is a significant part of their dietary fibre content. The composition and structure of chia nutlet mucilage are complex and contain several types of polymers. The fermentation of chia nutlet mucilage is poor, with only oligosaccharides being fermentable substrates. Grinding chia nutlets improves the fermentation and bioaccessibility of key nutrients, including dietary fibre.

Summary: The microstructure of legumes, particularly the protein matrix and cell wall of cotyledon cells, plays a crucial role in regulating starch digestion and glycemic responses. This study found that the protein matrix, in addition to the primary cell wall, acts as a secondary barrier to inhibit the accessibility of alpha-amylase to starch. Understanding the relationship between legume structure and starch digestion could inform the development of low glycemic index foods.

Perceived Usability of Tablet Crushers: Comparison of Devices by People with and without Limited Hand Functions

Hand-held twist-action crushers with an ergonomic grip received the highest usability scores among both groups. Hemicellulose-bacterial cellulose ribbon interactions affect the anisotropic mechanical behaviour of bacterial cellulose hydrogels

Summary: The aim of this study is to investigate the effect of hemicellulose-cellulose interactions on the anisotropic structure of the bacterial cellulose (BC) ribbon network. BC-arabinoxylan (BC-AX) and BC-mixed linkage glucan (BC-MLG) composites were prepared and their mechanical/rheological behavior was evaluated. The results showed that hemicelluloses had limited influence on the crystallinity of BC, but significantly affected the anisotropic behavior of BC hydrogels in compression and stress-relaxation tests.

Health literacy in Pakistani migrants in Australia-An emerging and neglected culturally and linguistically diverse community

Summary: This study investigated the health literacy of Pakistani migrants residing in Australia. The findings showed that while Pakistani respondents scored high in certain domains of health literacy, they scored low in others. Factors such as education, age, language proficiency, and residency status were associated with variations in health literacy among this community.

Composition and functional profiles of human faecal microbiota fermenting plant-based food particles are related to water-holding capacity more than particle size

Summary: This study investigated the effects of different particle sizes on microbial composition and functionality in plant-based foods through in vitro fermentation. The results showed that particle size, water-holding capacity, and pectin content were closely associated with microbial composition and fermentation outcomes. Particle size had little impact on microbial composition and functionality in wheat grain and apple samples, but it was related to alterations in the microbial composition and functional profiles of carrot samples.

Enzymatic arabinose depletion of wheat arabinoxylan regulates in vitro fermentation profiles and potential microbial degraders

Summary: This study investigated the fermentability of wheat arabinoxylan before and after hydrolysis by selective enzymes, and found that lower arabinose-to-xylose ratios and lower di-substitution levels contributed to slower substrate disappearance but higher microbial diversity. The structure of arabinoxylan also affected microbial beta diversity, with different genera associated with enzymatically-modified samples. Gas kinetics and end-product analyses showed that gas production rates depended on the ratio and substitution patterns, and faster fermentation rates resulted in different proportions of propionate, acetate, and butyrate. Overall, this study reveals the potential link between microbial arabinoxylan degraders and fermentation outcomes specific to the fine structures of arabinoxylan.

Summary: Lipid biomarkers, such as BHT isomers, can be used to trace bacterially mediated nitrogen and carbon cycle processes affecting greenhouse gas emissions. The source of BHT-34R in freshwater environments can be determined by its 813C value, and the presence of BHT-34R with low 813C values can be used as a novel tool to trace anammox in freshwater environments.
