Summary: This systematic review and meta-analysis evaluated the efficacy and safety of neoadjuvant PD-1/PD-L1 inhibitors combined with CTLA-4 inhibitors in malignant solid tumors. The results showed that the addition of CTLA-4 inhibitors did not significantly improve overall response rate and survival outcomes. However, there was an increased risk of grade 3-4 adverse events with the combination therapy. More large-scale and multicenter randomized controlled trials are needed to obtain more reliable results.

Dynamic assessment of treatment response through biomarkers like circulating PD-L1 and immune cell quantity, as well as consideration of host factors like microbiome and lifestyle, are important for integrating immunomodulating therapies into breast cancer management.

Summary: With the introduction of immunotherapies, there is new hope for cancer therapy, especially for cancers with poor prognoses like glioblastoma multiforme (GBM). However, these therapies are only effective in a small percentage of patients, highlighting the need for accurate predictors of response. In our study, we found that while TILs and PD-L1 expression are detectable in the majority of GBM samples, they do not significantly correlate with overall survival.

Summary: In this study, novel immune checkpoint inhibitors with enhanced potency were developed through modifications of an existing unnatural pentacyclic compound. These inhibitors showed the ability to suppress both CTLA-4 and PD-L1 gene expression and protein expression, making them the first reported dual small-molecule inhibitors of CTLA-4 and PD-L1.

Summary: This study aimed to determine the prognostic value of pulmonary hypertension (PH), right ventricular (RV) dysfunction, and tricuspid regurgitation (TR) on mortality after transcatheter mitral valve repair (TMVR). The results showed that patients with significant mitral regurgitation (MR) and concomitant PH, RV dysfunction, or TR had worse prognosis after TMVR. Careful assessment of these parameters should be done before making clinical decisions for TMVR.

Summary: A systematic review and meta-analysis found that fetuses with premature atrial contractions (PAC) have a 4-5 times higher risk of congenital heart defects (CHD) compared to the general population. The incidence of CHD was higher in the high-risk population. Advanced ultrasound examination is recommended for accurate diagnosis of PAC and exclusion of CHD, and weekly fetal heart-rate monitoring is advised to detect arrhythmia early and prevent cardiac failure.

This systematic review examines the evidence linking prenatal exposures to brain biomarkers associated with AD. The review identified 79 studies reporting on prenatal exposures such as alcohol, smoking, illicit drugs, and nutritional deficiencies, which were associated with smaller brain volumes. The findings highlight the importance of the prenatal environment in shaping the risk of neurodegenerative diseases in later life.
