Platelet-mediated shedding of NKG2D ligands impairs NK cell immune-surveillance of tumor cells

Platelets promote metastasis, among others by coating cancer cells traveling through the blood, which results in protection from NK cell immune-surveillance. The underlying mechanisms, however, remain to be fully elucidated. Here we report that platelet-coating reduces surface expression of NKG2D ligands, in particular MICA and MICB, on tumor cells, which was mirrored by enhanced release of their soluble ectodomains. Similar results were obtained upon exposure of tumor cells to platelet-releasate and can be attributed to the sheddases ADAM10 and ADAM17 that are detectable on the platelet surface and in releasate following activation and at higher levels on platelets of patients with metastasized lung cancer compared with healthy controls. Platelet-mediated NKG2DL-shedding in turn resulted in impaired induced self recognition by NK cells as revealed by diminished NKG2D-dependent lysis of tumor cells. Our results indicate that platelet-mediated NKG2DL-shedding may be involved in immune-evasion of (metastasizing) tumor cells from NK cell reactivity.

Summary: This study provides experimental evidence for the translational regulation of cancer-associated genes through uORFs, and demonstrates the frequent expression and occasional specific localization of uORF-derived peptides. Immunopeptidome analysis identified a set of naturally presented uORF-derived peptides, some of which are tumor-associated and capable of inducing multifunctional T cell responses. These findings suggest a potentially widespread function of uORFs and uPeptides in cancer biology.

Summary: Non-alcoholic steatohepatitis (NASH) negatively affects the efficacy of immune checkpoint inhibitor (ICI) therapy in hepatocellular carcinoma (HCC). This study reveals that NASH alters the metabolism, function, and motility of hepatic CD8+ T cells, leading to reduced responsiveness to anti-PD-1 treatment. The efficacy can be rescued by metformin treatment.

Summary: Using a large diverse cohort, this study found that the Omicron variant can bind to ACE2 more efficiently than inhibitory antibodies. Antibodies elicited by infection or immunization showed reduced binding capacities and ACE2 binding inhibition compared to wild type. Both Omicron variants are able to evade control by preexisting antibodies.

Increased soluble HLA in COVID-19 present a disease-related, diverse immunopeptidome associated with T cell immunity

Summary: Levels of soluble HLA molecules (sHLA) are significantly increased in COVID-19 patients and convalescent individuals, positively correlating with SARS-CoV-2-directed cellular immunity. Severe COVID-19 cases show reduced sHLA levels. The soluble immunopeptidome in COVID-19 patients exhibits increased diversity of HLA-presented peptides, and a SARS-CoV-2-derived peptide from the viral nucleoprotein is identified. Levels of sHLA directly correlate with the diversity of the soluble immunopeptidome.

Viral T-cell epitopes - Identification, characterization and clinical application

Summary: T-cell immunity is crucial for controlling viral infections, and it involves the CD4+ and CD8+ T cells. The viral T-cell epitopes, which are viral peptides presented by human leukocyte antigen (HLA) on virus-infected cells, are essential for T cell recognition of infected cells. Understanding viral T-cell epitopes not only offers a diagnostic tool to study protective T-cell responses in the human population, but also provides potential prophylactic and therapeutic options, such as vaccination and transfer of virus-specific T cells. These approaches have demonstrated effectiveness against various viral infections, especially in immunocompromised patients with insufficient antibody-based immune responses.
