Summary: This study used the NETSARC database to report the management and oncologic outcomes in adolescents and young adults (AYAs) patients with sarcoma in France. The study found that AYAs treated in reference sarcoma centers had improved locoregional recurrence-free survival and progression-free survival compared to those treated in non-reference centers. Therefore, referring AYAs patients with sarcoma to national sarcoma reference centers for comprehensive treatment is crucial.

Summary: This study aims to identify AYA sarcoma survivors with impaired social functioning (ISF) and determine the clinical and psychosocial factors associated with ISF. The study found that more than one-quarter of AYA sarcoma survivors experience ISF long after diagnosis. Unemployment and having to make lifestyle changes due to financial problems caused by physical conditions or medical treatment were associated with ISF, while better social support was associated with non-ISF. These results emphasize the importance of follow-up care that focuses not only on the disease but also on the psychological and social domains.

Specific support is necessary to maintain the quality of life (QOL) of AYA brain tumor patients, who may experience negative effects on neurological, neurocognitive, and endocrinological function, as well as on education, employment, and marriage opportunities. The biology underlying the distribution of tumors in AYAs may influence future clinical studies and molecular targeted therapy selection.

Target engagement is demonstrated in tumor tissue through elevated levels of the EYA3 substrate H2AX-pY142 upon loss of EYA3 or with Benzarone treatment.

Summary: This study analyzed the activation of receptor tyrosine kinase (RTK) pathway in Ewing sarcoma tumors and found that the tumors could be divided into two groups, one insensitive to IGF1R inhibition and the other sensitive to IGF1R or WEE1-targeted inhibitors. Primary and metastatic tumors also exhibited differences in IGF1R localization, replication stress levels, and inhibitor sensitivity. Combined inhibition of IGF1R and WEE1 led to tumor regression. These findings highlight the importance of using biopsy-derived predictive biomarkers at multiple stages of Ewing sarcoma disease management.
