Summary: This study designed chitosan nanoparticles containing miR-155-expressing plasmid and investigated their potential as an adjuvant to enhance immune responses. The prepared nanoparticles showed appropriate size and potential, and effectively protected the plasmid from degradation. The nanoparticles were able to downregulate the expression of miR-155 target genes and increase nitric oxide production, resulting in a shift of T cell responses towards Th1. These findings suggest that chitosan nanoparticles containing miR-155-expressing plasmid have the potential to significantly enhance immune responses.

Summary: Allergic contact dermatitis (ACD) is a type IV hypersensitivity mediated by Th1/Th17 immune response. A novel immunosuppressant, the DYRK1B inhibitor, was found to reduce ear inflammation and inhibit Th1 and Th17 cell differentiation while promoting Treg differentiation. This is potentially a promising novel agent for ACD treatment.

Summary: This study investigates the potential of micelles composed of polylactide-block-poly(N-acryloxysuccinimide-co-N-vinylpyrrolidone) as a delivery system for vaccines. The micelles were non-toxic and efficiently taken up by immune cells. Coupling of antigenic protein p24 on the micelles improved its antigenicity. In mouse immunization, the micelles showed comparable immune response intensity to gold standards and higher avidity index over time.

Summary: Developing adjuvant vaccines to combat multidrug-resistant Acinetobacter baumannii infections is a promising and cost-effective approach. In this study, a pDNA-CPG C274-adjuvant nano-vaccine was constructed and its immunogenicity and protection in mice were investigated. The vaccine demonstrated small size, positive charge, and spherical shape, and showed a continuous slow release pattern. It activated the TLR-9 pathway and produced increased amounts of total-IgG, IFN-gamma, and IL-1B in serum samples. Additionally, the vaccine provided potent protection against acute fatal A. baumannii challenge. These findings suggest that this nano-vaccine is a promising adjuvant approach for avoiding A. baumannii infection.

Summary: This study evaluated the preventative and therapeutic effects of a subunit vaccine against a virulent M.avium isolate causing pulmonary disease. Results showed that immunization with CFA+GLA-SE/CDG induced stronger CFA-specific Th1/Th17 immune responses and provided better protection in the lung and spleen. However, this vaccine was ineffective in reducing bacterial loads during treatment.

The study shows that the S-Trimer vaccine, when adjuvanted with SWE, induces strong antibody and cellular immune responses, providing neutralizing antibodies against the original and variant strains of SARS-CoV-2. Additionally, the SWE adjuvant allows for a lower vaccine dosage while maintaining effectiveness.

Summary: In silico chemogenomics approach was used to identify drugs or drug candidates with potential anti-MABSC activity by comparing the sequences of M. abscessus proteins with therapeutic targets. Six drugs including ezetimibe, furosemide, itraconazole, miconazole (MCZ), tamoxifen (TAM), and thiabendazole (THI) were selected for experimental validation. MCZ and TAM showed promising antibacterial activity against M. abscessus and can be used as molecular scaffolds for the development of new analogs.
