Occupational exposure to respirable crystalline silica and risk of autoimmune rheumatic diseases: a nationwide cohort study

Summary: Occupational exposure to respirable crystalline silica is associated with autoimmune rheumatic diseases, particularly systemic sclerosis and rheumatoid arthritis. The risk increases with higher cumulative exposure levels, indicating potential causal effects.

Adverse events of special interest in clinical trials of rheumatoid arthritis, psoriatic arthritis, ulcerative colitis and psoriasis with 37 066 patient-years of tofacitinib exposure

Summary: This study analyzed adverse events of special interest in tofacitinib clinical programs across four diseases, finding that infections were the most common adverse events in all diseases. The incidence rates for herpes zoster and serious infections were highest, and the adjusted mortality ratios weighted for country were relatively low across cohorts. The safety profile of tofacitinib was generally consistent across diseases and with longer term follow-up compared to previous analyses.

Neutrophil extracellular trap-associated carbamylation and histones trigger osteoclast formation in rheumatoid arthritis

Summary: Neutrophil infiltration into the synovial joint is a hallmark of rheumatoid arthritis (RA). This study found that carbamylation, a posttranslational modification, is linked to increased bone erosion in RA. The researchers demonstrated that neutrophil extracellular traps (NETs) and their carbamylated protein cargo directly promote bone destruction by inducing osteoclast formation.

Incidence and risk factors for herpes zoster in patients with rheumatoid arthritis receiving upadacitinib: a pooled analysis of six phase III clinical trials

Summary: In patients with RA, treatment with Upadacitinib (UPA) was associated with higher incidence and event rates of herpes zoster (HZ) compared to treatment with methotrexate (MTX) monotherapy or adalimumab (ADA) + MTX. The risk of HZ was also higher with the 30 mg dose of UPA compared to the 15 mg dose. Patients from Asia and those with a history of HZ may have an increased risk of HZ while receiving UPA.

Eggerthella lenta augments preclinical autoantibody production and metabolic shift mimicking senescence in arthritis

Summary: Although the exact cause of rheumatoid arthritis (RA) is still unknown, there is evidence to suggest a strong genetic predisposition and the presence of preclinical antibodies before symptoms manifest. This study investigates the impact of an expansion of Eggerthella lenta on the severity of RA using a humanized mouse model. The results show that mice colonized with E. lenta develop more severe disease when induced for arthritis, particularly in female mice. Furthermore, among RA patients, women tend to have a higher abundance of E. lenta. The expansion of E. lenta is associated with changes in immune responses, gut dysbiosis, and systemic inflammation.

Summary: This study identified a novel IL-17A/F-derived peptide and generated a new anti-IL-17 antibody that can effectively reverse the pro-inflammatory actions of IL-17A/F. The antibody showed less off-target effects compared to the current gold-standard biologic. Preclinical and clinical evidence demonstrated the high efficacy and therapeutic potency of the antibody in patients with IMIDs.

Summary: This study is the first to show an association between the rs2609255 variant of the FAM13A gene and interstitial lung disease (ILD) in Japanese patients with rheumatoid arthritis (RA), suggesting its involvement in the pathogenesis of usual interstitial pneumonia (UIP). Further studies on the function of rs2609255 are needed.

Summary: This study investigated the association between serum PBB-153 levels and autoimmune disorders. The results showed that increasing serum PBB-153 levels were associated with rheumatoid arthritis and neurological- and thyroid-related autoimmune disorders. The study also identified non-linear differences in this association by lifestage of exposure and sex.

Summary: This study found that in addition to occupational activities, women with rheumatoid arthritis (RA) may also be exposed to crystalline silica (SiO2) through cleaning activities and washing dusty clothes more than the general population. The study also found that high exposure to SiO2 may be associated with mediastinal lymphadenopathy.

Summary: By nano-formulating hydroxychloroquine and designing it as a macrophage-targeted nanotherapeutic, it is possible to achieve highly effective treatment for rheumatoid arthritis. This therapeutic approach can induce repolarization of macrophages towards an anti-inflammatory phenotype, effectively reducing inflammation, and has shown significant therapeutic effects in mouse models.

Summary: In this study, a inflammation-specific environment activated methotrexate-encapsulated nanomedicine (MTX@NPs) was developed for the treatment of rheumatoid arthritis (RA). It effectively accumulated in inflamed joints and released methotrexate in the specific RA microenvironment. MTX@NPs demonstrated a high therapeutic efficacy for RA with minimal systemic toxicity by suppressing inflammation response and improving lymphatic functions.

Antiphospholipid antibody-mediated NK cell cytotoxicity

Summary: Antiphospholipid syndrome (APS) is an autoimmune thrombophilia characterized by thrombosis and obstetric complications in the presence of antiphospholipid antibodies (aPL). Our study aimed to investigate the role of natural killer (NK) cells in early pregnancy loss in APS. We found that aPL can recruit antibody-dependent cellular cytotoxicity (ADCC) of NK cells, with differences observed between aPL containing antiB2GPI domain 1 (anti-beta 2GPI-D1) antibodies (aPL+/D1+) and those that do not (aPL+/D1-). This suggests that NK cells may play a role in APS-related obstetric complications.
