Lactoferrin Efficiently Counteracts the Inflammation-Induced Changes of the Iron Homeostasis System in Macrophages

Human lactoferrin (hLf), an 80-kDa multifunctional iron-binding cationic glycoprotein, is constitutively secreted by exocrine glands and by neutrophils during inflammation. hLf is recognized as a key element in the host immune defense system. The in vitro and in vivo experiments are carried out with bovine Lf (bLf), which shares high sequence homology and identical functions with hLf, including anti-inflammatory activity. Here, in pure M1 human macrophages, obtained by stimulation with a mixture of 10 pg/ml LPS and 20 ng/ml IFN-gamma, as well as in a more heterogeneous macrophage population, challenged with high-dose of LPS (1 pg/ml), the effect of bLf on the expression of the main proteins involved in iron and inflammatory homeostasis, namely ferroportin (Fpn), membrane-bound ceruloplasmin (Cp), cytosolic ferritin (Ftn), transferrin receptor 1, and cytokines has been investigated. The increase of IL-6 and IL-1 beta cytokines, following the inflammatory treatments, is associated with both upregulation of cytosolic Ftn and downregulation of Fpn, membrane-bound Cp, and transferrin receptor 1. All these changes take part into intracellular iron overload, a very unsafe condition leading in vivo to higher host susceptibility to infections as well as iron deficiency in the blood and anemia of inflammation. It is, therefore, of utmost importance to counteract the persistence of the inflammatory status to rebalance iron levels between tissues/secretions and blood. Moreover, levels of the antiinflammatory cytokine IL-10 were increased in cells treated with high doses of LPS. Conversely, IL-10 decreased when the LPS/IFN-7 mix was used, suggesting that only the inflammation triggered by LPS high doses can switch on an anti-inflammatory response in our macrophagic model. Here, we demonstrate that bLf, when included in the culture medium, significantly reduced IL-6 and IL-1 beta production and efficiently prevented the changes of Fpn, membrane-bound Cp, cytosolic Ftn, and transferrin receptor 1 in pure M1 macrophages, as well as in the more heterogeneous macrophage population. In addition, the decrease of IL-10 induced by the LPS/IFN-7 mix was counteracted by bovine lactoferrin. Several drugs capable of modulating mac rophagic phenotypes are emerging as attractive molecules for treating inflammation, and in this sense, bovine lactoferrin is no exception.

Lactoferrin as Immune-Enhancement Strategy for SARS-CoV-2 Infection in Alzheimer's Disease Patients

Summary: COVID-19 poses a greater risk to older adults and those with dementia. Lactoferrin may have antiviral and anti-inflammatory properties against SARS-CoV-2, improving immune system responses and potentially serving as a treatment for COVID-19. Reduced levels of lactoferrin in saliva may increase the susceptibility of Alzheimer's disease patients to COVID-19. Bovine lactoferrin may be a promising preventive therapeutic tool to minimize COVID-19 risk in dementia or Alzheimer's disease patients.

An observational survey on patients with recurrent cystitis demonstrated a significant reduction in episodes after oral administration of bLf.

An overview on in vitro and in vivo antiviral activity of lactoferrin: its efficacy against SARS-CoV-2 infection

Summary: This article reviews the antiviral activity of lactoferrin (Lf) against various types of viruses, including SARS-CoV-2. The study found that oral administration of Lf can counteract COVID-19 pathogenesis in the early stage of SARS-CoV-2 infection and have effects on various physiological processes such as inflammation, iron ion regulation, and coagulation balance. Lactoferrin: from the structure to the functional orchestration of iron homeostasis

Summary: Iron is an essential transition metal with crucial biological functions, but it also poses challenges for organisms due to its insolubility and formation of toxic radicals. Organisms have evolved extracellular carrier proteins to acquire and transport iron. Dysregulation of iron-proteins has been implicated in various pathological conditions, yet no resolutive therapy has been found. This review highlights the efficacy of Lactoferrin, a member of the transferrin family, in managing iron disorders associated with different pathologies.

Experimental-theoretical study of laccase as a detoxifier of aflatoxins

Summary: We investigated the detoxification of aflatoxins, fungal carcinogenic food contaminants, by laccase. Experimental comparison of two similar aflatoxins (AFB(1) and AFG(2)) showed significant differences in laccase-mediated detoxification. A multi-scale modeling approach identified substrate-specific changes needed to improve laccase's detoxification capability, and a density functional theory-based model determined the amino acid residues affecting laccase's affinity for aflatoxins. The study concluded that AFB(1) is more challenging to degrade, while AFG(2) is easier due to its lack of side products and favorable binding dynamics, highlighting opportunities to optimize laccase for aflatoxin degradation.

Iron Saturation Drives Lactoferrin Effects on Oxidative Stress and Neurotoxicity Induced by HIV-1 Tat

Summary: The study examines the effect of bovine lactoferrin (bLf) on oxidative stress and neurotoxicity in human astroglial cells infected with Human Immunodeficiency Virus (HIV-1). The results show that both forms of bLf, nat-bLf and holo-bLf, enhance the host antioxidant response and reduce reactive oxygen species-mediated damage in astrocytes. The study also demonstrates that bLf restores the physiological internalization of transferrin receptor 1 and exacerbates Tat-induced excitotoxicity on human neuronal cells.

Genetic Incorporation of Dansylalanine in Human Ferroportin to Probe the Alternating Access Mechanism of Iron Transport

In this study, a fluorescent amino acid (DA) was introduced into specific positions of human Fpn to investigate conformational changes during metal translocation. The results indicate metal-induced conformational transitions, providing insights into the alternating access mechanism of Fpn and advancing our understanding of iron transport.
