The genetic variants of CHRNA7 and CHRFAM7A can alter cytokine responses and are involved in immune responses. The CHRFAM7A gene may have a more pronounced role in regulating inflammatory processes in humans.

Summary: This study found that human ASM cells express alpha 7nAChR receptors, with higher levels in asthmatics and smokers, and further increased by exposure to nicotine, cigarette smoke, or pro-inflammatory cytokines TNF alpha and IL-13. These receptors may play a potential role in asthma pathophysiology through signaling pathways such as NF kappa B, AP-1, and CREB.

Summary: In this study, it was found that the alpha 7 nicotinic acetylcholine receptor agonist GTS-21 can alleviate inflammatory and metabolic abnormalities in a rat model of ischemic stroke. It exerts its neuroprotective effects by inhibiting the expression of pro-inflammatory cytokines and promoting anti-inflammatory cytokines, as well as improving brain infarction size and motor coordination ability.

Summary: During melanoma metastasis, the innate inflammatory response in the metastatic microenvironment of the sentinel lymph node (sLN) was characterized. Macrophages in the subcapsular sinus (SS) produced protumoral IL1a after recognizing tumoral antigens. The blocking of IL1a reduced metastatic spread and had a synergistic antimetastatic effect when combined with STAT3 inhibition.

Summary: This study investigated the role of non-neuronal cholinergic system in osteoarthritis (OA). The results showed that the cholinergic system, through alpha 7nAchR, can counteract the effects of inflammation on osteoblasts. Deletion of alpha 7nAchR may limit cartilage degradation in age-related OA. Therefore, enhancing the cholinergic system could be a potential therapeutic target, although it may depend on CHRFAM7A expression.

Summary: The alpha 7 and alpha 4 beta 2* nicotinic acetylcholine receptors are the most abundant nAChRs in the mammalian brain and are targeted in drug discovery for brain disorders. A C(10) variant of cytisine has been developed as a partial agonist of alpha 4 beta 2 nAChR. Molecular dynamic simulations and experiments revealed the structural basis for the selectivity of 10-methylcytisine for alpha 7 and alpha 4 beta 2 nAChRs, providing insights into agonist binding and receptor dynamics.

Summary: The study found that PNU-282987 significantly reduced goblet cell hyperplasia, eosinophil infiltration, and ILC2 numbers in the airways following IL-33 or AA challenges. In vitro experiments showed a decrease in GATA3 and Ki67 expression in isolated lung ILC2s after treatment with PNU-282987 or GTS-21. Additionally, PNU-282987 treatment resulted in decreased IKK and NF-kappa B phosphorylation compared to GTS-21 in ILC2s.

Summary: The study confirms the key role of the hippocampus in the mechanism of hypoxic preconditioning, as well as the importance of alpha 7nAChR activation in hypoxic preconditioning. It was found in the experiment that the activation of alpha 7nAChRs is not involved in the cholinergic signaling initiated by HBH or DMSO via any route of administration.
