Summary: In this study, a high-throughput targeted proteomic method was used to systematically investigate the temporal responses of the small GTPase proteome during osteogenic differentiation of human embryonic stem cells. The method showed high quantification accuracy, reproducibility, and throughput. The results revealed altered expression of various small GTPases during osteogenic differentiation, with KRAS playing a previously unrecognized role in regulating extracellular matrix accumulation for mineralization.

Summary: In this study, the authors found that Glutaredoxin3(GLRX3) is an important redox-regulating molecule associated with nucleus pulposus (NP) cell senescence and disc degeneration. They developed GLRX3(+) mesenchymal stem cell-derived extracellular vehicles (EVs-GLRX3) that enhanced antioxidant defense and prevented senescence cascade expansion. They also proposed a disc tissue-like biopolymer-based supramolecular hydrogel for delivering EVs-GLRX3 to treat disc degeneration. The EVs-GLRX3-loaded hydrogel attenuated mitochondrial damage, alleviated NP senescence, and restored ECM deposition.

Summary: A new strategy was developed to reinforce microbial fuel cell performance by engineering synthetic extracellular matrix with cytochrome fusion. The engineered strain showed significantly higher voltage and power density output in MFC compared to the wild type, suggesting the potential of this approach in improving EET and MFC efficiency. This work opens up new possibilities for exoelectrogen design and engineering through ECM manipulation.
