Mfsd2a (Major Facilitator Superfamily Domain Containing 2a) Attenuates Intracerebral Hemorrhage-Induced Blood-Brain Barrier Disruption by Inhibiting Vesicular Transcytosis

Background-Blood-brain barrier (BBB) disruption aggravates brain injury induced by intracerebral hemorrhage (ICH); however, the mechanisms of BBB damage caused by ICH remain elusive. Mfsd2a (major facilitator superfamily domain containing 2a) has been known to play an essential role in BBB formation and function. In this study, we investigated the role and underlying mechanisms of Mfsd2a in BBB permeability regulation after ICH. Methods and Results-Using ICH models, we found that Mfsd2a protein expression in perihematomal brain tissues was significantly decreased after ICH. Knockdown and knockout of Mfsd2a in mice markedly increased BBB permeability, neurological deficit score, and brain water contents after ICH, and these were rescued by overexpressing Mfsd2a in perihematomas. Moreover, we found that Mfsd2a regulation of BBB permeability after ICH correlated with changes in vesicle number. Expression profiling of tight junction proteins showed no differences in Mfsd2a knockdown, Mfsd2a knockout, and Mfsd2a overexpression mice. However, using electron microscopy following ICH, we observed a significant increase in pinocytotic vesicle number in Mfsd2a knockout mice and decreased the number of pinocytotic vesicles in mouse brains with Mfsd2a overexpression. Finally, using multiple reaction monitoring, we screened out 3 vesicle trafficking-related proteins (Srgap2, Stx7, and Sec22b) from 31 vesicle trafficking-related proteins that were markedly upregulated in Mfsd2a knockout mice compared with controls after ICH. Conclusions-In summary, our results suggest that Mfsd2a may protect against BBB injury by inhibiting vesicular transcytosis following ICH.

The results showed that clinicians would generally feel comfortable skipping IVT if there was reasonable comparability between IVT+EVT and EVT alone, with no more than 3% of patients being unable to achieve functional independence at 90 days due to skipping IVT.

Astrocyte Reprogramming in Stroke: Opportunities and Challenges

Redistribution of Histone Marks on Inflammatory Genes Associated With Intracerebral Hemorrhage-Induced Acute Brain Injury in Aging Rats

Summary: This study investigates the contribution of histone mark redistribution to the age-induced decline of endogenous neuroprotection in an intracerebral hemorrhage-induced acute brain injury rat model. The findings reveal that neuroinflammation-associated genes are upregulated in ICH rat brains regardless of age, with interferon-gamma response genes activated in both age groups. Anti-IFN-gamma treatment effectively reduces acute brain injury in older rats. Additionally, histone modifications, particularly H3K4me3 and H3K27me3, are redistributed in neuroinflammation-associated genes in both age groups and are correlated with gene expression. These findings highlight the potential of targeting histone modifications as a therapeutic approach for age-related diseases.

miR-129-5p targets FEZ1/SCOC/ULK1/NBR1 complex to restore neuronal function in mice with post-stroke depression

Metabolomics profiling to characterize cerebral ischemia-reperfusion injury in mice

Summary: This study investigated the metabolic alterations during ischemia-reperfusion injury using metabolomic analysis. A total of 82 differentially expressed metabolites were identified, with some significantly elevated in the reperfusion group. Metabolomics pathway analysis revealed the importance of glycerophospholipid metabolism, linoleic acid metabolism, pyrimidine metabolism, and galactose metabolism in the response to cerebral ischemia-reperfusion. This study provides important insights into the metabolic changes induced by cerebral ischemia-reperfusion.

Compared with final successful reperfusion, FPE also independently contributed to favorable outcomes.

Effect of endovascular treatment within 6 hours for acute basilar artery occlusion

Summary: The study aimed to investigate the efficacy and safety of combining endovascular treatment (EVT) with standard medical treatment (SMT) in Chinese patients with acute basilar artery occlusion (BAO) within 6 hours of occlusion. Patients were divided into SMT-alone group or EVT+SMT group. Results showed that EVT was associated with a higher rate of favorable functional outcome and a lower proportion of deaths. Predictors of favorable outcome in the EVT+SMT group included younger age, lower baseline National Institutes of Health Stroke Scale score, higher baseline posterior circulation Alberta Stroke Program Early CT Score, absence of diabetes mellitus, and modified Thrombolysis in Cerebral Infarction scores 2b-3.
