Prevalence, Host Range, and Comparative Genomic Analysis of Temperate Ochrobactrum Phage

Ochrobactrum and Brucella are closely related bacteria that populate different habitats and differ in their pathogenic properties. Only little is known about mobile genetic elements in these genera which might be important for survival and virulence. Previous studies on Brucella lysogeny indicated that active phages are rare in this genus. To gain insight into the presence and nature of prophages in Ochrobactrum, temperate phages were isolated from various species and characterized in detail. In silico analyses disclosed numerous prophages in published Ochrobactrum genomes. Induction experiments showed that Ochrobactrum prophages can be induced by various stress factors and that some strains released phage particles even under non-induced conditions. Sixty percent of lysates prepared from 125 strains revealed lytic activity. The host range and DNA similarities of 19 phages belonging to the families Myoviridae, Siphoviridae, or Podoviridae were determined suggesting that they are highly diverse. Some phages showed relationship to the temperate Brucella inopinata phage BiPB01. The genomic sequences of the myovirus POA1180 (41,655 bp) and podovirus POI1126 (60,065 bp) were analyzed. Phage POA1180 is very similar to a prophage recently identified in a Brucella strain isolated from an exotic frog. The POA1180 genome contains genes which may confer resistance to chromate and the ability to take up sulfate. Phage POI1126 is related to podoviruses of Sinorhizobium meliloti (PCB5), Erwinia pyrifoliae (Pep14), and Burkholderia cenocepacia (BcepIL02) and almost identical to an unnamed plasmid of the Ochrobactrum intermedium strain LMG 3301. Further experiments revealed that the POI1126 prophage indeed replicates as an extrachromosomal element. The data demonstrate for the first time that active prophages are common in Ochrobactrum and suggest that atypical brucellae also may be a reservoir for temperate phages.

The diversity of resistance genes was highest in Klebsiella spp. from wastewater treatment plants, with no carbapenemase-encoding genes detected.

The results showed that the combined method was effective in detecting and isolating E. coli or Salmonella spp. strains harboring different mcr genes, and could potentially be used as a harmonized protocol for screening mcr genes in food-producing animals and food products in Europe.

Summary: This study investigated the occurrence of fosfomycin-resistant Enterobacterales, particularly E. coli, in city wastewater treatment plants and hospital sewage in Hatay, Turkey. The isolates were characterized for their clonal relatedness, resistomes, and mobile genetic elements, revealing the potential for the dissemination of fosfomycin resistance. Plasmid-mediated, transmissible FosR in E. coli from wastewater poses a foreseeable threat to One-Health.

Characterization of Temperate LPS-Binding Bordetella avium Phages That Lack Superinfection Immunity

Summary: In this study, seven Bordetella avium phages were isolated and characterized, showing broad host range and strong bacteriolytic activity. These phages used lipopolysaccharides as host receptors and exhibited a myovirus structure. Although phenotypically thought to be lytic, these phages were found to undergo a lysogenic phase, but infection did not confer stable host superinfection immunity. These findings are important for potential phage therapy against avian bordetellosis.

Global colistin use: a review of the emergence of resistant Enterobacterales and the impact on their genetic basis

Summary: The global rise of MDR and XDR Enterobacterales in human medicine has led to the reintroduction of colistin as a last-resort drug. However, the use of colistin in veterinary medicine has resulted in a steadily increasing prevalence of resistant isolates in livestock and meat-based food. The transmission of resistant isolates from animals to humans, acquiring resistance through food, and exposure to colistin in clinical settings have contributed to the increased prevalence of colistin-resistant Enterobacterales in humans. This study focuses on the understanding of chromosomal-mediated resistance mechanisms in Enterobacterales, providing an overview of the genes involved in colistin resistance and the global situation of colistin-resistant Enterobacterales.
