Summary: This study reveals that under hypoxic conditions, the alternative splicing of MBD2 is suppressed, leading to the production of MBD2a which promotes breast cancer metastasis. Clinical data show a significant correlation between the expression of MBD2a and MBD2c with the invasiveness of breast cancer, indicating their opposing roles in regulating metastasis in this disease.

Summary: The oncogene MYC promotes cancer progression by down-regulating N6-methyladenosine (m(6)A) modification preferentially in transcripts of MYC-repressed genes (MRGs). This alteration leads to the activation of ALKBH5 expression and reduction of m(6)A levels in specific MRGs, controlling their translation through the m(6)A reader YTHDF3. Ultimately, inhibiting ALKBH5 or overexpressing SPI1 or PHF12 effectively suppresses the growth of MYC-deregulated B-cell lymphomas.

Summary: This study identified KDELR2 as a novel target of HDAC3, with aberrant expression indicating poor prognosis in breast cancer patients. The correlation between HDAC3 and KDELR2 in tumor tissues was confirmed. It was found that HDAC3 transactivated KDELR2 via CREB1, promoting cell cycle progression and tumorigenesis. The HDAC3-KDELR2 axis could be a potential therapeutic target for breast cancer.

Summary: Metformin increases sensitivity of hepatocarcinoma cells to methotrexate by suppressing DHFR expression, blocking nucleotide metabolism, and inhibiting cell cycle progression and tumorigenesis. It represses DHFR transcriptionally via E2F4 and promotes lysosomal degradation of DHFR protein. Metformin enhances patient-derived hepatocarcinoma organoids response to MTX without toxicity to normal liver organoids, suggesting a potential therapeutically targetable vulnerability in hepatocarcinoma.

Aberrant activation of ENO1/YAP1/PLCB1 and decreased HPGD expression in liver cancer samples suggest a potential correlation between ENO1-regulated AA metabolism and cancer development, highlighting the therapeutic potential of aspirin.
