Variety of RNAs in Peripheral Blood Cells, Plasma, and Plasma Fractions

Human peripheral blood contains RNA in cells and in extracellular membrane vesicles, microvesicles and exosomes, as well as in cell-free ribonucleoproteins. Circulating mRNAs and noncoding RNAs, being internalized, possess the ability to modulate vital processes in recipient cells. In this study, with SOLiD sequencing technology, we performed identification, classification, and quantification of RNAs from blood fractions: cells, plasma, plasma vesicles pelleted at 16,000.. and 160,000.., and vesicle-depleted plasma supernatant of healthy donors and non-small cell lung cancer (NSCLC) patients. It was determined that 16,000.. blood plasma vesicles were enriched with cell-free mitochondria and with a set of mitochondrial RNAs. The variable RNA set of blood plasma 160,000.. pellets reflected the prominent contribution of U1, U5, and U6 small nuclear RNAs' fragments and at the same time was characterized by a remarkable depletion of small nucleolar RNAs. Besides microRNAs, the variety of fragments of mRNAs and snoRNAs dominated in the set of circulating RNAs differentially expressed in blood fractions of NSCLC patients. Taken together, our data emphasize that not only extracellular microRNAs but also circulating fragments of messenger and small nuclear/nucleolar RNAs represent prominent classes of circulating regulatory ncRNAs as well as promising circulating biomarkers for the development of disease diagnostic approaches.

Chemical Modifications Influence the Number of siRNA Molecules Adsorbed on Gold Nanoparticles and the Efficiency of Downregulation of a Target Protein

Summary: Small interfering RNAs (siRNAs) are powerful tools for suppressing protein synthesis in the cell, but they face challenges such as low resistance to nucleases and inability to enter target cells. In this study, researchers designed multilayer nanoconstructs (MLNCs) with gold nanoparticle cores carrying chemically modified siRNAs. By introducing chemical modifications, the efficiency of siRNA adsorption onto the gold core was increased. The stability of modified siRNA on the gold core was also tested, and MLNCs with the most effective siRNA cargo were selected. These MLNCs showed increased biological activity compared to control MLNCs.

Chloroquine Enhances Death in Lung Adenocarcinoma A549 Cells Exposed to Cold Atmospheric Plasma Jet

Summary: Cold atmospheric plasma (CAP) is a widely studied approach for treating malignant neoplasms. This study determined the optimal discharge regime for direct CAP treatment of cancer cells and found that CAP weakly activated pro-survival autophagy in irradiated cells. The addition of the autophagy inhibitor chloroquine (CQ) promoted CAP-dependent cell death.

Summary: We have studied the possibility of obtaining appropriate preparations of EVs from whole-body extract of holothuria Eupentacta fraudatrix using centrifugation and ultracentrifugation. However, the preparations were heavily polluted and subsequent purification didn't increase vesicle concentration to a necessary level. To confirm the presence of EVs, including exosomes, in holothurian cells, we used transmission electron microscopy and found late endosomes in the coelom epithelium and parenchyma cells of visceral organs.

Template-Assisted Assembly of Hybrid DNA/RNA Nanostructures Using Branched Oligodeoxy- and Oligoribonucleotides

Summary: A template-assisted assembly approach to a C24 fullerene-like double-stranded DNA polyhedral shell is proposed. The assembly employed a supramolecular oligonucleotide dendrimer as a 3D template that was obtained via the hybridization of siRNA strands and a single-stranded DNA oligonucleotide joined to three- or four-way branched junctions. The article discusses the use of a photolabile protecting group to prevent premature auto-hybridization of the self-complementary oligonucleotides, as well as the incorporation of cleavable linkers to guide the post-assembly disconnection of the shell. The study also explores the modification of siRNA strands to enhance enzymatic stability.

Changes in the Ultrastructure of Staphylococcus aureus Cells Make It Possible to Identify and Analyze the Injuring Effects of Ciprofloxacin, Polycationic Amphiphile and Their Hybrid

Summary: Understanding the relationship between the effects of antibacterial compounds and their chemical structure is crucial for the purposeful development of synthetic antibacterial compounds. In this study, we examined the ultrastructural changes in S. aureus cells caused by the polycationic amphiphile DL(4)12, ciprofloxacin, and their hybrid DL(5)Cip6. Our findings revealed that DL(4)12 damaged the bacterial cell wall and disrupted the cytoplasm, while ciprofloxacin altered the cell membrane and cytoplasm and inhibited bacterial division. The ultrastructural characteristics of S. aureus cells damaged by the hybrid differed from those under ciprofloxacin or DL(4)12 action. Furthermore, we observed clear differences in the effects of these compounds on the morphology of S. aureus biofilm.

The Molecular Basis for Selectivity of the Cytotoxic Response of Lung Adenocarcinoma Cells to Cold Atmospheric Plasma

Summary: This study uncovers the molecular mechanisms behind the selective death of tumor cells under cold atmospheric plasma (CAP) exposure. The response to CAP treatment was analyzed in lung adenocarcinoma cells and lung-fibroblast cells, revealing common trends in both cell lines but with variable amplitude in the tumor cells. CAP treatment resulted in DNA damage, cell-cycle arrest, and ER lumens dysfunction in the tumor cells. These findings suggest that nuclear stress and ER stress contribute to the differential sensitivity of tumor and healthy cells to CAP exposure.

Human Blood Extracellular Vesicles Activate Transcription of NF-kB-Dependent Genes in A549 Lung Adenocarcinoma Cells

Summary: Extracellular vesicles (EVs) are heterogeneous in size and composition, and changes in their RNA sets can be used for diagnostic and therapeutic approaches. This study obtained EVs from the blood of healthy donors and analyzed their RNA composition. The results suggest that EVs can trigger the NF-kB signaling pathway in lung adenocarcinoma cells, leading to the transcription of genes involved in cytokines, adhesion molecules, cell cycle regulation, and cell survival.

MicroRNA and mRNA Expression Changes in Glioblastoma Cells Cultivated under Conditions of Neurosphere Formation

Summary: The formation of neurospheres in GBM cells is accompanied by changes in the expression of miRNAs and their target mRNAs. Upregulated miRNAs are associated with decreased levels of their target mRNAs, while downregulated miRNAs are associated with increased levels of their target mRNAs. These miRNAs and their target mRNAs may play important roles in the regulation of malignant growth and invasion in GBM.
