Inactivation of Fam20B in the dental epithelium of mice leads to supernumerary incisors

Tooth formation is tightly regulated by epithelial-mesenchymal interactions via hierarchic cascades of signaling molecules. The glycosaminoglycan (GAG) chains covalently attached to the core protein of proteoglycans (PGs) provide docking sites for signaling molecules and their receptors during the morphogenesis of tissues and organs. Although PGs are believed to play important roles in tooth formation, little is known about their exact functions in this developmental process and the relevant molecular basis. Family with sequence similarity member 20-B (FAM20B) is a newly identified kinase that phosphorylates the xylose in the common linkage region connecting the GAG with the protein core of PGs. The phosphorylation of xylose is essential for elongation of the common linkage region and the subsequent GAG assembly. In this study, we generated a Fam20B-floxed allele in mice and found that inactivating Fam20B in the dental epithelium leads to supernumerary maxillary and mandibular incisors. This finding highlights the pivotal role of PGs in tooth morphogenesis and opens a new window for understanding the regulatory mechanism of PG-mediated signaling cascades during tooth formation.

An in situ hybridization study of syndecan family during the late stages of developing mouse molar tooth germ

Summary: The expression patterns of syndecan-1, 2, 3, and 4 mRNAs during late tooth germ formation were investigated. Syndecan-1 mRNA was mainly expressed in specific tooth germ regions and its expression in dental epithelium increased during the postnatal periods. Syndecan-3 mRNA was found to be involved in dentinogenesis, while syndecan-2 showed enhanced expression with hard tissue formation. Syndecan-4 exhibited similar expression patterns to syndecan-3.

RANKL inhibition halts lesion progression and promotes bone remineralization in mice with fibrous dysplasia

Summary: Fibrous dysplasia (FD) is a rare bone disease caused by GNAS mutation, and there is currently no cure available. Inhibition of RANKL expression has been identified as a potential therapy for FD. This study evaluated the effects of a small-molecule RANKL inhibitor (A52676293) on a transgenic FD mouse model, and found that it effectively suppressed osteoclastogenesis and halted FD progression. The results suggest that RANKL inhibition could be an effective treatment for FD.

Methods to assess tooth gingival thickness and diagnose gingival phenotypes: A systematic review

Summary: In this study, a heterostructured coating was created on a porous bioinert polyetheretherketone implant. The heterostructured coating significantly promotes cytocompatibility and osteogenic differentiation through multiple mechanisms and effectively kills bacteria through photothermal/photodynamic effects and photo-induced accelerated ion release. The results confirmed the excellent osteogenicity and osseointegration of the heterostructured orthopedic implant, as well as its photoswitchable disinfection controlled by NIR light.

Effect of occlusal coverage depths on the precision of 3D-printed orthognathic surgical splints

Summary: The study proposed a standardized protocol for 3D-printed splints and assessed the precision of splints with different occlusal coverage depths. Results showed that deviations were more evident in the vertical dimension and pitch rotation, increasing as the occlusal coverage depth increased. Splints with 2-mm and 3-mm occlusal coverage depths are recommended for precision relative to clinical acceptability.

In this study, a novel peptide MY-1 is designed and synthesized, and nano-hydroxyapatite (nHA) is used as the carrier for sustained release of MY-1. The results show that sustained release of MY-1 regulates the synthesis and secretion of ECM, promoting cell migration and osteogenic differentiation, which benefits bone regeneration.

Optimal cuspal coverage of ceramic restorations using CAD/CAM: Biomechanical characteristic analysis by 3D finite element analysis and in vitro investigation

Summary: This study evaluated the stress distribution patterns, resistance to fracture, and failure modes of endodontically treated molars restored with different cuspal coverage options. The results showed that onlay restorations with whole functional cuspal coverage provided comparable effects to coverage of all cusps in terms of stress distribution, fracture resistance, and mechanical performance in high occlusal areas.
