Proteomic Analysis of Pemphigus Autoantibodies Indicates a Larger, More Diverse, and More Dynamic Repertoire than Determined by B Cell Genetics

In autoantibody-mediated diseases such as pemphigus, serum antibodies lead to disease. Genetic analysis of B cells has allowed characterization of antibody repertoires in such diseases but would be complemented by proteomic analysis of serum autoantibodies. Here, we show using proteomic analysis that the serum autoantibody repertoire in pemphigus is much more polyclonal than that found by genetic studies of B cells. In addition, many B cells encode pemphigus autoantibodies that are not secreted into the serum. Heavy chain variable gene usage of serum autoantibodies is not shared among patients, implying targeting of the coded proteins will not be a useful therapeutic strategy. Analysis of autoantibodies in individual patients over several years indicates that many antibody clones persist but the proportion of each changes. These studies indicate a dynamic and diverse autoantibody response not revealed by genetic studies and explain why similar overall autoantibody titers may give variable disease activity.

Summary: There is a bidirectional association between bullous pemphigoid (BP) and atopic dermatitis (AD) and allergic rhinitis (AR). A history of AD and AR increases the risk of developing BP, while BP patients are at an increased risk of subsequent AD. BP patients with comorbid AD and AR are more likely to be managed with drugs and corticosteroids, and have reduced all-cause mortality.

Summary: The study reveals that many human tumors have reduced levels of Mic60, an essential scaffold of mitochondrial structure, which leads to the disruption of mitochondrial integrity. Surprisingly, these tumors show decreased cell proliferation, resistance to cell death, and activation of a gene expression program related to innate immunity and cytokine/chemokine signaling. This process induces epithelial-mesenchymal transition, tumor cell movement, and promotes metastatic dissemination.

Summary: This study found that the oxidant peroxynitrite (PNT) can alter the profile of MHC-I bound peptides presented on tumor cells. Only cytotoxic cells targeting PNT-resistant peptides have an anti-tumor effect, while those targeting PNT-sensitive peptides are ineffective. Targeting PNT in mice reduces tumor cell resistance to cytotoxic cells. Melanoma patients with low PNT activity in their tumors respond better to immunotherapy.

Updates on the immunopathology and genomics of severe cutaneous adverse drug reactions

Summary: Severe cutaneous adverse reactions (SCARs), such as Stevens-Johnson syndrome (SJS), toxic epidermal necrolysis (TEN), and drug reaction with eosinophilia and systemic symptoms (DRESS)/drug-induced hypersensitivity syndrome (DIHS), have significant impact on health and hinder drug development. Understanding the association between HLA class I alleles and SCARs has helped with prevention efforts and shed light on the immunopathogenesis. However, there is still much unknown about the specific risk alleles and antigens involved. Identifying the MHC-restricted drug-reactive peptides and genetic/environmental risk factors will be crucial for predicting individual and drug-level risks and developing targeted therapeutics.

Parkin ubiquitination of Kindlin-2 enables mitochondria-associated metastasis suppression

Summary: In this study, it is found that the altered ubiquitination ligase Parkin in Parkinson's disease forms a complex with the regulator of cell motility Kindlin-2 at mitochondria of tumor cells. Parkin ubiquitinates Kindlin-2 leading to its proteasomal degradation, which inhibits tumor cell interactions with the extracellular matrix, migration, and invasion. However, Parkin does not affect tumor cell proliferation, cell cycle transitions, or apoptosis.

This study found that CARM1 promotes monounsaturated fatty acid synthesis and targeting the fatty acid reprogramming pathway can be a potential therapeutic strategy for CARM1-expressing ovarian cancers.
