Summary: In this study, the researchers investigated the factors that regulate compartment specific mRNA translation in human cells. They found that the glycolytic enzyme Pyruvate Kinase M (PKM) interacts with polysomes and influences mRNA translation, and this interaction is regulated by ADP levels. Further experiments indicated that PKM binds to mRNA sequences downstream of lysine- and glutamate-enriched tracts and causes translational stalling near these sequences. The recruitment of PKM to polysomes depends on poly-ADP ribosylation activity, suggesting a link between cellular metabolism and mRNA translation.

Summary: This study reveals the role of SMAD3 in oncogene-induced senescence and its conversion to malignant transformation. SMAD3, activated by high-level transforming growth factor B1 (TGF-B1) following oncogene Ras overactivation in lung epithelial cells, induces cellular senescence through the repression of cell cycle-promoting genes. Interestingly, SMAD3 also has an oncogenic effect and facilitates oncogene Ras-driven malignant transformation. Furthermore, the study identifies the potential tumor suppressor ATOH8 as a binding partner of SMAD3 and shows that its loss accelerates oncogene Ras-driven lung tumorigenesis.

The epigenetic remodelers ARID1A and NSD2 have been identified as key regulators in promoting IFN-I expression by enhancing chromatin accessibility of IRF3 at the Ifn-I promoters. This study provides insight into the crosstalk between chromatin remodeling, histone modification, and transcription factors in the epigenetic regulation of antiviral innate immunity.

Summary: Obesity is a common chronic metabolic disease, and inducing apoptosis in preadipocytes and adipocytes is a potential strategy for its treatment. Celastrol was found to induce apoptosis in preadipocytes through a mitochondrial mediated pathway. It was further discovered that celastrol inhibits autophagy by preventing the fusion of autophagosomes and lysosomes, leading to cell apoptosis.

Summary: Tetracycline antibiotics targeting the mitoribosome were found to protect against sepsis by perturbing the electron transport chain and reducing tissue damage in the lung, while increasing fatty acid oxidation and glucocorticoid sensitivity in the liver. This study demonstrates that mitochondrial protein synthesis inhibition leading to ETC perturbation is a mechanism for inducing disease tolerance beyond the antibacterial activity of these antibiotics.

Summary: The study found that constant darkness rearing in rats can affect synaptic transmission and function between the retinohypothalamic tract (RHT) and suprachiasmatic nucleus (SCN) during early postnatal ages but lead to synaptic response strengthening at later ages. The results suggest that the environmental conditions induce a compensatory mechanism in the glutamatergic signaling to enhance synchronization to light at adult ages, and that the synaptic properties of RHT terminals during postnatal development are not critically influenced by light exposure.
