Summary: In response to colonization by rhizobia bacteria, legumes have developed a complex, long-distance signaling pathway to regulate nodulation in their roots and shoots. We measured the transcriptional response to rhizobia treatment in Medicago truncatula and used the GeneShift tool to detect temporal shifts in gene expression. This study identified known and novel genes that are dynamically regulated during early nodulation and validated the findings using an independent RNA-Seq study.

Summary: Plant epigenetic regulations play important roles in transposable element silencing, developmental processes, and responses to the environment. Specifically, DNA methylation modifications are involved in these processes. The development of root nodules, which are important for nitrogen fixation, is a complex process with various regulatory mechanisms. In this study, laser capture microdissection and RNA-sequencing techniques were used to identify genes activated in the nodule differentiation and nitrogen fixation zones. MtDME, which is upregulated in the differentiation zone, was found to be required for nodule development. Additionally, differentially methylated regions were identified in the nodule. CRISPR-Cas9 mutagenesis of MtDRM2 further demonstrated the importance of RdDM in CHH hypermethylation and nodule development. A model of DNA methylation dynamics during nodule development was proposed.

The findings provide detailed insights into the PcG family in M. truncatula and offer potential for further research in legumes.

Summary: This study used single-nucleus RNA sequencing to characterize the roots of Medicago truncatula after nod factor treatment at different time points. The results showed drastic global gene expression reprogramming in the epidermis and cortex at 30 min, with most changes restored at 6 h. Plant defence response genes were activated at 30 min and subsequently suppressed at 6 h in non-meristem cells. Highly expressed flavonoid synthase genes required for rhizobia recruitment were found in cortical cells at 30 min. The analysis of a gene module enriched for symbiotic nitrogen fixation genes revealed that MtFER and LYK3 shared similar responses to symbiotic signals. Furthermore, it was found that LYK3 could phosphorylate MtFER and participate in rhizobial symbiosis.

Summary: Growth Regulatory Factors (GRF) are plant-specific transcription factors that play critical roles in plant growth and development as well as plant tolerance against stress. In this study, 16 GRF genes were identified from the genomes of Medicago truncatula and Medicago sativa, and their sequences and expression patterns were analyzed. The results showed that these GRF genes are important for plant growth and stress tolerance, particularly in response to salt and drought stress.

Summary: Iron deficiency in plants leads to excessive production of reactive oxygen species, affecting plant growth and development depending on the genotype's tolerance to iron deficiency. The study showed that iron deficiency decreases photosynthetic capacity and biomass production, with tolerant genotypes maintaining iron content and increasing copper concentration. Under iron deficiency, there was a significant increase in H2O2, but tolerant genotypes showed a significant increase in SOD activity.

Summary: Class 2 KNOX transcription factors play a role in promoting cell differentiation in plants and have different functions in Arabidopsis and Medicago truncatula. In Medicago truncatula, KNOX2 genes control symbiotic root nodule formation. We conducted a study to investigate the role of MtKNOX3-like genes in drought response. Our results showed that knockdown of these genes in transgenic M. truncatula lines resulted in decreased tolerance to drought conditions, suggesting their involvement in drought response pathways through the regulation of proline degradation.

Summary: In the genetic study of durum wheat in Canada, QTL mapping was conducted to identify loci associated with grain protein concentration (GPC). Several major and minor QTL were discovered on different chromosomes, with some being consistently detected across multiple years and showing potential for improving both GPC and grain yield. The results of traditional and conditional mapping provide valuable insights for the development of high GPC and high yielding durum cultivars.
