Summary: Acute liver injury (ALI) is a severe liver disease characterized by sudden hepatocyte necrosis and liver function deterioration. This study develops hepatocyte-targeting self-assembling nanoparticles encapsulating an organic Selenium compound, which effectively removes reactive oxygen species (ROS) and protects hepatocyte viability. The nanoparticles, functionalized with a liver-targeting ligand, exhibit enhanced hepatocyte uptake and successfully reduce liver damage in mouse models of ALI induced by acetaminophen or carbon tetrachloride.

Summary: A novel conjugation strategy was explored to make potent antibiotics more selective by accumulating the drug and releasing it triggered by infection. Bacterial targeting was achieved using a modified fragment of a human antimicrobial peptide, and the release of the drug was limited to infection-related situations through a linker cleaved by neutrophil elastase. The antibacterial efficacy of the conjugates was successfully demonstrated in co-cultures of human neutrophils and Escherichia coli.

Summary: A novel light activatable micron-sized liposomal drug carrier was developed for repetitive drug release based on NIR pulsed laser cycles, achieved through the photothermal effect of gold nanorods attached to liposomes. The liposomes demonstrated repeatability in drug release due to their reversible nature, releasing drug via lipid bilayer melting under laser irradiation and quickly regaining original structure afterward. The study also evaluated the long-term stability, drug encapsulation efficiency, and drug-releasing efficiency of these liposomes, suggesting their potential as long-term drug delivery vehicles.

Summary: Photochemical internalisation (PCI) is a method that uses sub-lethal photodynamic therapy (PDT) to control cytosolic drug delivery, activating a photosensitiser with non-ionising visible light. This study examines the efficacy of PCI on ovarian cancer tumouroids using a porphyrin photosensitiser and a cytotoxin (Saporin), showing that PCI effectively decreases cancer cell viability within the central cancer mass and the surrounding stroma. Compared to treatment with Saporin or PDT alone, PCI significantly reduces stromal invasion by cancer cells. Tumouroids are suitable models for studying multi-cellular events after pharmacological assault.
