E6AP promotes prostate cancer by reducing p27 expression

Prostate cancer (PC) is the most common cancer in men. Elevated levels of E3 ligase, E6-Associated Protein (E6AP) were previously linked to PC, consistent with increased protein expression in a subset of PC patients. In cancers, irregular E3 ligase activity drives proteasomal degradation of tumor suppressor proteins. Accordingly, E3 ligase inhibitors define a rational therapy to restore tumor suppression. The relevant tumor suppressors targeted by E6AP in PC are yet to be fully identified. In this study we show that p27, a key cell cycle regulator, is a target of E6AP in PC. Down regulation of E6AP increases p27 expression and enhances its nuclear accumulation in PC. We demonstrate that E6AP regulates p27 expression by inhibiting its transcription in an E2F1-dependent manner. Concomitant knockdown of E6AP and p27 partially restores PC cell growth, supporting the contribution of p27 to the overall effect of E6AP on prostate tumorigenesis. Overall, we unravelled the E6AP-p27 axis as a new promoter of PC, exposing an attractive target for therapy through the restoration of tumor suppression.

This study discusses the surgical treatment of mandibular ameloblastomas and explores the use of new technologies in reconstruction.

Summary: Grading of breast phyllodes tumors (PTs) varies due to differences in weighting of grading criteria by pathologists. Key diagnostic criteria include increased stromal cellularity, stromal overgrowth, increased stromal mitoses, stromal atypia, stromal fronding, irregular tumor borders, and/or lesional heterogeneity. Clinical and radiological findings, as well as age, also play important roles in determining the diagnosis.

Identifying recurrences and metastasis after ductal carcinoma in situ (DCIS) of the breast

Summary: Ductal carcinoma in situ (DCIS) is a non-invasive breast tumor with the potential to progress to invasive ductal carcinoma (IDC). The current inability to accurately predict the invasive potential of DCIS leads to both over-treatment and under-treatment in patients. Better understanding of the molecular features of DCIS could improve treatment decision-making and avoid unnecessary side-effects. This review summarizes the current knowledge of DCIS and discusses future research directions.

Microenvironmental Factors in Oral Cavity Squamous Cell Carcinoma Undergoing Surgery: Correlation with Diffusion Kurtosis Imaging and Dynamic Contrast-Enhanced MRI

Summary: In this study, the correlation between imaging parameters obtained from magnetic resonance imaging (MRI) and pathological factors in oral cavity squamous cell carcinomas (OSCCs) was investigated. The results showed that pathological factors such as inflammatory infiltrate, tumor grading, and desmoplastic reaction affected the MRI parameters. Significant relationships were found between DKI parameters and tumor-infiltrating lymphocytes (TILs). Differences in DCE-MRI parameters were also observed according to tumor grading. These findings contribute to a better understanding of OSCCs and can be used for diagnostic and prognostic purposes.

Development of a somatic variant registry in a National Cancer Center: towards Molecular Real World Data preparedness

Summary: The biomedical research field is advancing towards developing clinical trials and translational projects based on real-world evidence. To make this transition feasible, clinical centers need to work towards data accessibility and interoperability. This task is particularly challenging when applied to genomics, which has become routine through amplicon-based next-generation sequencing panels. These experiments produce numerous features per patient, and their results are often stored in static clinical reports, making critical information inaccessible to automated access and federated search consortia. This study presents a reanalysis of 4620 solid tumor sequencing samples in different histology settings and describes the bioinformatics and data engineering processes employed to create a somatic variant registry capable of dealing with the large biotechnological variability of routine genomics profiling.

Spatial transcriptomics and the anatomical pathologist: Molecular meets morphology

Summary: In recent years, molecular findings have been incorporated into routine diagnostic practice of anatomical pathology, revolutionizing the field. Spatial transcriptomics, a group of technologies that provide transcriptome-wide expression profiling while preserving spatial origin of the data, allows analysis of highly specific areas within a tissue section and correlation of expression data with morphology, tissue type, and location relative to other structures. Some spatial transcriptomics methods are now compatible with formalin-fixed paraffin-embedded tissue (FFPE), potentially leading to their incorporation in routine anatomical pathology practice.
