Beneficial Effect of Voluntary Exercise on Experimental Colitis in Mice Fed a High-Fat Diet: The Role of Irisin, Adiponectin and Proinflammatory Biomarkers

Inflammatory bowel diseases (IBDs) are a heterogeneous group of disorders exhibited by two major phenotypic forms: Crohn's disease and ulcerative colitis. Although the aetiology of IBD is unknown, several factors coming from the adipose tissue and skeletal muscles, such as cytokines, adipokines and myokines, were suggested in the pathogenesis of ulcerative colitis; however, it has not been extensively studied whether voluntary exercise can ameliorate that disorder. We explored the effect of moderate exercise (i.e., voluntary wheel running) on the disease activity index (DAI), colonic blood flow (CBF), plasma irisin and adiponectin levels and real-time PCR expression of proinflammatory markers in mesenteric fat in mice with 2,4,6-trinitrobenzenesulfonic acid (TNBS) colitis fed a high-fat diet (HFD) compared to those on a standard chow diet (SD). Macroscopic and microscopic colitis in sedentary SD mice was accompanied by a significant fall in CBF, some increase in colonic tissue weight and a significant increase in the plasma levels of tumour necrosis factor-alpha (TNF-alpha), IL-6, monocyte chemotactic protein 1 (MCP-1) and IL-13 (p < 0.05). In sedentary HFD mice, colonic lesions were aggravated, colonic tissue weight increased and the plasma TNF-alpha, IL-6, MCP-1, IL-1 beta and leptin levels significantly increased. Simultaneously, a significant decrease in the plasma irisin and adiponectin levels was observed in comparison with SD mice (p < 0.05). Exercise significantly decreased macroscopic and microscopic colitis, substantially increased CBF and attenuated the plasma TNF-alpha, IL-6, MCP-1, IL-1 beta and leptin levels while raising the plasma irisin and the plasma and WAT concentrations of adiponectin in HFD mice (p < 0.05). We conclude that: (1) experimental colitis is exacerbated in HFD mice, possibly due to a fall in colonic microcirculation and an increase in the plasma and mesenteric fat content of proinflammatory biomarkers; and (2) voluntary physical activity can attenuate the severity of colonic damage in mice fed a HFD through the release of protective irisin and restoration of plasma adiponectin.

Inhaled silica nanoparticles exacerbate atherosclerosis through skewing macrophage polarization towards M1 phenotype

Summary: This study investigated the effect of silica and ferric oxide nanoparticles on the development of atherosclerosis, finding that inhaled silica nanoparticles increased atherosclerotic lesions, while exposure to ferric oxide nanoparticles caused significant changes in plaque composition. Both nanoparticles altered the phenotype of T lymphocytes in the spleen.

Profile of circulating extracellular vesicles microRNA correlates with the disease activity in granulomatosis with polyangiitis

Summary: This study characterized the profile of microRNAs trafficked by extracellular vesicles (EVs) in patients with granulomatosis with polyangiitis (GPA). Four differentially expressed microRNAs were identified in EVs from GPA patients, with miR-223-3p being the best discriminator between GPA patients and healthy controls. MiR-664a-3p was able to discriminate between active and remission phases of GPA. The correct classification rate based on multivariate discriminant analysis was 92% for acute phase and 85% for all study participants. Pathway analysis revealed potential target genes of these microRNAs, involved in focal adhesion, mTOR signaling, and neutrophil extracellular traps formation. Two microRNAs positively correlated with disease activity were found to be involved in neutrophil extracellular traps formation and apoptosis inhibition.

Summary: In this study, it was found that obesity and sedentary lifestyle worsened colitis in mice, while supplementation with intestinal alkaline phosphatase (IAP) and moderate physical activity attenuated the severity of colitis in obese mice. This improvement may be achieved through downregulation of the intestinal cytokine/chemokine network and oxidative stress, modulation of gut microbiota, and improvement of muscle strength.

Chronic Vulvar Pain and Health-Related Quality of Life in Women with Vulvodynia

Summary: This study examined the severity of vulvar pain and its impact on quality of life in a group of Polish women. It was found that vulvodynia mainly affects young, highly educated, and unmarried women, divorcees, and widows. The disease significantly reduces quality of life, mainly due to difficulties in daily activities and decreased sexual satisfaction. The severity of pain is negatively correlated with quality of life. Continued research in this area is recommended.

The Two-Track Investigation of Fibronectin Binding Protein A of Staphylococcus aureus from Bovine Mastitis as a Potential Candidate for Immunodiagnosis: A Pilot Study

Summary: Bovine mastitis is a common and costly disease for dairy cattle breeders worldwide. This study aimed to detect and characterize the fibronectin binding protein A (FnBPA) of Staphylococcus aureus (SA) in milk samples from cows with mastitis. Bioinformatic analysis identified FnBPA as a potential biomarker for bovine SA. Immunoblotting confirmed the presence and immunoreactivity of FnBPA, suggesting it could be a valuable biomarker for mastitis immunodiagnostics.

Summary: This review assessed the impact of antenatal perineal massage (APM) on perinatal perineal injuries and the development of pelvic pain and other complications in postpartum women. The findings showed that APM can reduce the risk of perineal injuries and pain during childbirth, as well as decrease the risk of fecal and gas incontinence in the postpartum period.
