Summary: This study identified the baseline high eosinophil count as a potential biomarker for predicting the clinical efficacy of CAR-T cell therapy in B-NHL patients. A preclinical model demonstrated that depletion of eosinophils could impair the intratumoral infiltration and antitumor efficacy of CAR-T cells.

Summary: This study revealed diminished T-cell infiltration and increased T-cell receptor immune repertoire diversity in clear cell renal cell cancer (ccRCC). Furthermore, Vimentin expression in ccRCC tissue was positively correlated with the presence of positive T-cell markers. The immune repertoire sequencing results showed increased diversity in the T-cell receptor repertoire in ccRCC.

Summary: This study investigates the impact of cGAS-STING expression and the presence of cancer-associated fibroblasts (CAFs) on immune cell infiltration in pancreatic ductal adenocarcinoma (PDAC). The results show that PDAC cases with co-expression of cGAS and STING have favorable survival and exhibit cytotoxic CD8 + T cell infiltrations. However, the presence of CAFs impedes immune cell infiltration.

Summary: The study demonstrates that enhancing CAR T cell therapy targeting solid tumors with STING agonists, specifically DMXAA, can greatly improve tumor control by promoting CAR T cell trafficking and persistence in the tumor microenvironment. The single-cell RNA sequencing results show that DMXAA generates a chemokine milieu that facilitates CART cell recruitment and modulates the immunosuppressive TME.

Summary: This study investigates the impact of transcriptomic changes on the killing ability of effector cytotoxic T lymphocytes (CTLs). The results show that while transcription is required for the expression of cytokines/chemokines and transcriptional machinery, it is relatively robust to transcription blockade for cytotoxic protein expression and cytolytic activity. Additionally, the study reveals a cell-intrinsic transcriptional requirement for infiltration.

Summary: This study found that the Twist2 stromal lineage cells play a crucial role in the growth and maintenance of intestinal stem cells (ISCs) and their ability to regenerate villi. Additionally, Acta2(+) cells also contribute to ISC regeneration. These findings suggest that different cellular niches are required for ISC-mediated villus homeostasis and regeneration.

Summary: An injectable hydrogel was designed to reinforce cancer immunotherapy by regulating CXCL10 degradation to amplify tumor T cell infiltration and inactivating PD-L1 to eliminate the PD-L1-governed immune evasion.

Summary: The study developed biocompatible immune cell-engaging particles (ICEp) for efficient and tunable protein loading, aiming to improve the safety of immunotherapy. Injection of micrometre-sized ICEp intratumorally activates CAR T cells, leading to local tumor clearance while protecting uninjected tumors. Control of immune cell activation can enhance the efficiency of cell-based therapies and reduce associated risks.

Summary: The study found that PDAC tumor tissue had richer and more diverse IG and TCR infiltration compared to normal pancreatic tissue. Specific IG clonotypes could help differentiate samples with better prognosis, while a larger TCR infiltration was associated with patients with a history of diabetes.

Summary: In this study, the significance of CD8(+) T cell-related markers in patients with KIRC was comprehensively analyzed. A 6-gene risk scoring model specific to TIL-CD8T was constructed based on multi-platform analyses. Different prognosis of patients with the same or similar immune checkpoint gene levels could be distinguished by TIL-CD8 sig score.

Summary: Research suggests that genetics, immune reactions, and the joint-gut axis play major roles in the etiology of ankylosing spondylitis (AS). While B cells have been traditionally understudied in AS pathogenesis, increasing evidence points to their involvement, with disturbances in B cell populations and the formation of antibodies seen in AS patients.

Summary: Immune checkpoint blockades have shown breakthroughs in certain types of cancer, but the response rate is low. The underlying mechanisms and efficient biomarkers to predict immunotherapy resistances are still unclear. This study found that BATF2 inhibits the expression of programmed death-ligand 1 in cancer cells by suppressing the PI3K-AKT pathway. BATF2 promotes antitumor immune response and can serve as an efficient biomarker to predict diagnosis, prognosis, and immunotherapy efficacy in patients with non-small cell lung cancer.

Summary: The study developed a novel tetravalent antibody F4L5.13, which can promote the expression of junction proteins on the surface of retinal endothelial cells and restore the function of the blood-retina-barrier in retinal development. Additionally, F4L5.13 can treat oxygen-induced retinopathy models characterized by abnormal neovascularization, demonstrating a new therapeutic strategy for diseases with abnormal angiogenesis and barrier dysfunction.
