Exosomal cargo including microRNA regulates sensory neuron to macrophage communication after nerve trauma

Following peripheral axon injury, dysregulation of non-coding microRNAs (miRs) occurs in dorsal root ganglia (DRG) sensory neurons. Here we show that DRG neuron cell bodies release extracellular vesicles, including exosomes containing miRs, upon activity. We demonstrate that miR-21-5p is released in the exosomal fraction of cultured DRG following capsaicin activation of TRPV1 receptors. Pure sensory neuron-derived exosomes released by capsaicin are readily phagocytosed by macrophages in which an increase in miR-21-5p expression promotes a pro-inflammatory phenotype. After nerve injury in mice, miR-21-5p is upregulated in DRG neurons and both intrathecal delivery of a miR-21-5p antagomir and conditional deletion of miR-21 in sensory neurons reduce neuropathic hypersensitivity as well as the extent of inflammatory macrophage recruitment in the DRG. We suggest that upregulation and release of miR-21 contribute to sensory neuron-macrophage communication after damage to the peripheral nerve.

Sexually Dimorphic Immune and Neuroimmune Changes Following Peripheral Nerve Injury in Mice: Novel Insights for Gender Medicine

Summary: This study identified gender differences in immune responses to neuropathic pain, with females showing higher infiltration of T cells in nerves and males showing higher infiltration of macrophages. Additionally, cytokine and chemokine levels were differently regulated in blood and nerve lysates from female mice compared to male mice. These findings provide insights into gender-associated inflammatory profiles in neuropathy and could contribute to the development of differential biomarkers and personalized medicine.

Municipal wastewater viral pollution in Saudi Arabia: effect of hot climate on COVID-19 disease spreading

Summary: This study investigated the spread of COVID-19 through wastewater and the efficiency of sewage treatment in removing the virus. The results showed that sewage treatment was effective in removing SARS-CoV-2 viral RNA. However, the viral RNA load in wastewater was relatively low, indicating that sewage may only preserve viral RNA for a short time in a hot climate. Additionally, there was no significant correlation between hospital admissions and viral RNA in sewage.

The results show that TNF-alpha and LPS, either alone or in combination, can induce the maturation of DCs in vitro. However, the absence of both TNF-alpha and LPS leads to an immature state of DCs and induces the expression of TIM-3.

Summary: The present study aimed to investigate the potential neuroprotective effect of Pioglitazone in PTZ kindled seizure in mice. Treatment with Pioglitazone demonstrated a significant decrease in NGF, seizure score, m-TOR, GLUT-1 and GLUT-3 in comparison to the PTZ group, suggesting that Pioglitazone provides its neuroprotective effect through modulating m-TOR signaling, glucose metabolism and NGF levels.

Summary: A new method was developed to create nanoparticles from freeze-and air-dried mangosteen peel, and the effects of these particles on the growth and protein formation of various gram-positive foodborne pathogenic bacteria were studied. The results showed that the freeze-dried nanoparticles had higher antioxidant activity compared to the air-dried ones, and they could inhibit the protein formation of bacteria.
