The IKK/NF-κB signaling pathway requires Morgana to drive breast cancer metastasis

NF-kappa B is a transcription factor involved in the regulation of multiple physiological and pathological cellular processes, including inflammation, cell survival, proliferation, and cancer cell metastasis. NF-kappa B is frequently hyperactivated in several cancers, including triplenegative breast cancer. Here we show that NF-kappa B activation in breast cancer cells depends on the presence of the CHORDC1 gene product Morgana, a previously unknown component of the IKK complex and essential for I kappa B alpha substrate recognition. Morgana silencing blocks metastasis formation in breast cancer mouse models and this phenotype is reverted by I kappa B alpha downregulation. High Morgana expression levels in cancer cells decrease recruitment of natural killer cells in the first phases of tumor growth and induce the expression of cytokines able to attract neutrophils in the primary tumor, as well as in the pre-metastatic lungs, fueling cancer metastasis. In accordance, high Morgana levels positively correlate with NF-kappa B target gene expression and poor prognosis in human patients.

Summary: Long non-coding RNAs (lncRNAs) are widely expressed in the human brain, and may play a role in normal brain development and the molecular pathology of neurodevelopmental disorders. However, their involvement in intellectual disability (ID) and neurodevelopmental disorders is still poorly understood, and only a few genomic alterations have been causally linked to the disease endophenotype. Further investigation into the non-coding genome may uncover novel pathogenic mechanisms and translational opportunities.

Single-cell atlas of the aging mouse colon

Summary: Little is known about the course of COVID-19 and the antibody response to infection or vaccination in patients with hairy-cell leukaemia (HCL). In this study of 58 HCL cases, 37 unvaccinated patients developed COVID-19 with a generally favorable outcome, while active leukemia, older age, and comorbidities were associated with a worse course. Seroconversion consistently developed after infection (11 cases) and vaccination (28 cases), except in patients recently treated with anti-CD20 or venetoclax therapy. Vaccination appeared to protect against severe COVID-19 in patients with breakthrough infection.

Shedding Light on the Pathogenesis of Splanchnic Vein Thrombosis

Summary: Splanchnic vein thrombosis is a rare but potentially life-threatening manifestation of venous thromboembolism that poses challenges in terms of pathology and treatment. It is commonly associated with liver cirrhosis, but can also be caused by myeloproliferative disorders, gastroenterological cancers, abdominal infections, and thrombophilia. Some cases of splanchnic vein thrombosis remain idiopathic. In this review, we discuss the mechanisms of splanchnic vein thrombosis, including new insights on the role of clonal hematopoiesis in the development of idiopathic SVT, which have important implications for treatment.

NR2F1 shapes mitochondria in the mouse brain, providing new insights into Bosch-Boonstra-Schaaf optic atrophy syndrome

Summary: The nuclear receptor NR2F1 is a strong transcriptional regulator in neural cells. Mutations in the NR2F1 gene cause BBSOAS, a rare neurodevelopmental disorder characterized by vision impairment, intellectual disability, and autistic traits. In this study, the researchers identified a set of nuclear-encoded mitochondrial genes as potential targets under direct NR2F1 control. They demonstrated that NR2F1 loss of function in mice resulted in reduced mitochondrial mass, fragmentation, and downregulation of key mitochondrial proteins in newborn neurons, impairing their generation, survival, and integration. Similar dysregulation was also observed in a BBSOAS mouse model. These findings highlight the role of NR2F1 in the mitochondrial gene expression network in neurons and suggest a connection between mitochondrial dysfunction and BBSOAS pathogenesis.
