Identification of NCCRP1 as an epigenetically regulated tumor suppressor and biomarker for malignant phenotypes of squamous cell carcinoma of the esophagus

The poor prognosis and increasing incidence of esophageal squamous cell carcinoma (ESCC) highlight the need for identification of novel ESCC-associated molecular events to improve the diagnosis, and treatment of this disease. Non-specific cytotoxic cell receptor protein 1 (NCCRP1) was reported to be abundantly expressed in human squamous epithelium and to be involved in cell proliferation; however, the role of NCCRP1 in ESCC remains unclear. To elucidate the oncological roles of NCCRP1 in ESCC, NCCRP1 expression, DNA methylation, and copy numbers were analyzed in ESCC cell lines. Nine ESCC cell lines demonstrated different NCCRP1 mRNA expression levels and all exhibited hypermethylation of the NCCRP1 promoter, but no copy number loss. Additionally, NCCRP1 expression was determined in 213 surgically resected esophageal tissue samples. NCCRP1 mRNA expression levels were reduced in ESCC tissues compared with corresponding non-cancerous adjacent tissues in 204 (95.8%) patients. Patients in the low NCCRP1 expression group tended to have a higher recurrence rate and a shorter overall survival time compared with those in the high NCCRP1 expression group. Additionally, multivariate analysis revealed that low NCCRP1 expression was an independent prognostic factor (hazard ratio, 1.75; 95% confidence interval, 1.08-2.87; P=0.022). The findings of the current study indicate that NCCRP1 acts as a putative tumor suppressor that is inactivated through promoter hypermethylation, and serves as a promising biomarker to predict postoperative prognosis in ESCC.

A liquid biopsy signature for predicting early recurrence in patients with gastric cancer

Summary: We established a miRNA-based liquid biopsy signature that can reliably predict early recurrence in gastric cancer patients by analyzing miRNA expression profiling. Additionally, a model combining the miRNA signature, microsatellite instability (MSI) status, and tumor size exhibited better predictive performance and was successfully validated in another independent cohort. Finally, we translated the miRNA signature into a liquid biopsy assay and demonstrated its independence as a predictor for early recurrence.

Risk score for predicting death from other causes after curative gastrectomy for gastric cancer

Summary: This study aimed to build a risk score for predicting deaths from other causes after gastrectomy in gastric cancer patients. Nine characteristics were identified as risk factors for poor survival, and patients were classified into Low-, Intermediate-, and High-risk groups based on their risk scores. The 5-year survival rates were significantly different among the three risk groups, highlighting the usefulness of this risk score for predicting deaths from other causes after curative gastrectomy.

Efficacy and feasibility of scheduled intravenous acetaminophen administration after pancreatoduodenectomy: a propensity score-matched study

Summary: This study aimed to investigate the efficacy and safety of routine intravenous administration of acetaminophen after highly invasive hepatobiliary pancreatic surgery, particularly pancreatoduodenectomy. The analgesic effects and adverse events of scheduled intravenous administration of acetaminophen were evaluated in 179 patients who underwent open pancreatoduodenectomy. The results showed that scheduled intravenous administration of acetaminophen did not affect postoperative liver function and had a good analgesic effect after pancreatoduodenectomy.

Targeting Pancreatic Cancer with Novel Plumbagin Derivatives: Design, Synthesis, Molecular Mechanism, In Vitro and In Vivo Evaluation

Summary: Pancreatic tumors in an austere tumor microenvironment activate adaptive pathways to tolerate nutrient starvation and promote aggressive malignancy. Plumbagin, a plant-derived compound, has shown cytotoxicity against pancreatic cancer cells in nutrient-deprived conditions. 2-(cyclohexylmethyl)-plumbagin was identified as the most promising compound that inhibits cancer cell survival under nutrient-deprived conditions by targeting the PI3K/Akt/mTOR pathways. In vivo studies using xenograft mouse models confirmed the effectiveness of this compound in inhibiting tumor growth. Plumbagin derivatives, such as 2-(cyclohexylmethyl)-plumbagin, hold great potential for the development of anticancer drugs targeting the tumor's austerity microenvironment.

Nicolaioidesin C: An Antiausterity Agent Shows Promising Antitumor Activity in a Pancreatic Cancer Xenograft Mouse Model

Summary: Pancreatic tumors are often characterized by hypoxia and severe nutrient deprivation, making anticancer drugs targeting rapidly proliferating cells ineffective. The search for novel agents that can eliminate cancer cells' adaptations to nutrition starvation, known as antiausterity agents, is promising. The natural product (+)-nicolaioidesin C (Nic-C) has shown potent antiausterity activity against pancreatic cancer cells in vitro, and its racemic form (synthesized Nic-C) inhibited tumor growth and signaling pathways in a human pancreatic cancer xenograft model. Metabolomic profiling suggests Nic-C affects amino acid and sphingolipid metabolism.
