Activation of the c-Jun NH2-terminal kinase pathway by coronavirus infectious bronchitis virus promotes apoptosis independently of c-Jun

Mitogen-activated protein kinases (MAPKs) are conserved protein kinases that regulate a variety of important cellular signaling pathways. Among them, c-Jun N-terminal kinases (JNK) are known to be activated by various environmental stresses including virus infections. Previously, activation of the JNK pathway has been detected in cells infected with several coronaviruses. However, detailed characterization of the pathway as well as its implication in host-virus interactions has not been fully investigated. Here we report that the JNK pathway was activated in cells infected with the avian coronavirus infectious bronchitis virus (IBV). Of the two known upstream MAPK kinases (MKK), MKK7, but not MKK4, was shown to be responsible for IBV-induced JNK activation. Moreover, knockdown and overexpression experiments demonstrated that JNK served as a pro-apoptotic protein during IBV infection. Interestingly, pro-apoptotic activity of JNK was not mediated via c-Jun, but involved modulation of the anti-apoptotic protein B-cell lymphoma 2 (Bcl2). Taken together, JNK constitutes an important aspect of coronavirus-host interaction, along with other MAPKs.

Activation of the MKK3-p38-MK2-ZFP36 Axis by Coronavirus Infection Restricts the Upregulation of AU-Rich Element-Containing Transcripts in Proinflammatory Responses

Summary: This study reveals that coronavirus infection induces the expression of ZFP36, an anti-inflammatory protein, and promotes the degradation of transcripts coding for IL-6 and IL-8, as well as other proteins related to inflammation. The p38 MAP kinase and its upstream kinase MKK3, as well as downstream kinase MK2, play a regulatory role in the upregulation of ZFP36 during coronavirus infection. This MKK3-p38-MK2-ZFP36 axis could be a potential therapeutic target for severe coronavirus infections.

The Methyltransferase HemK Regulates the Virulence and Nutrient Utilization of the Phytopathogenic Bacterium Xanthomonas citri Subsp. citri

Summary: Citrus canker, caused by Xanthomonas citri subsp. citri, is a devastating disease that affects fruit quality and yield. This study found that a protein called HemK plays a crucial role in regulating the virulence traits of Xcc. Deletion of the hemK gene resulted in decreased motility, biofilm formation, extracellular enzymes, and polysaccharide production, as well as increased sensitivity to oxidative stress and high temperatures. HemK was also found to be essential for the virulence of Xcc on citrus plants and for the hypersensitive response on a non-host plant.

Transcriptomic analysis reveals crucial regulatory roles of immediate-early response genes and related signaling pathways in coronavirus infectious bronchitis virus infection

Summary: This study presents the transcriptomic profile of cells infected with gammacoronavirus infectious bronchitis virus (IBV) and reveals the differential expression of host genes and their related pathways. The study identifies 1162 differentially expressed genes, mainly enriched in MAPK and Wnt signaling pathways, in IBV-infected human non-small cell lung carcinoma cells (H1299 cells). The validation of induced transcripts in IBV-infected cells demonstrates the accuracy and reliability of the transcriptomic data. This study provides a reliable transcriptomic profile of host genes and pathways regulated by coronavirus infection.

Avian IRF1 and IRF7 Play Overlapping and Distinct Roles in Regulating IFN-Dependent and -Independent Antiviral Responses to Duck Tembusu Virus Infection

The upregulation of EGR1 has differential effects on viral replication, apoptosis, and antiviral response.

Summary: Disruption of the cell cycle is a common strategy used by many viruses to create a favorable cellular microenvironment for their replication. In this study, we demonstrate that the gamma-coronavirus infectious bronchitis virus (IBV) nonstructural protein 12 (nsp12) interacts with the p50 regulatory subunit of DNA polymerase delta (pol delta), as well as with nsp13, to induce cell cycle arrest in the S phase. Interestingly, nsp12 from severe acute respiratory syndrome coronavirus (SARS-CoV) and SARS-CoV-2 also interacted with p50. These findings highlight the importance of these interactions in coronavirus replication and virus-host interaction, and suggest potential targets for antiviral intervention.

Summary: Coronavirus infection induces a variety of cellular antiviral responses either dependent on or independent of type I interferons (IFNs). Our previous studies revealed the differential induction of three IFN-stimulated genes (ISGs), IRF1, ISG15 and ISG20, by gammacoronavirus infectious bronchitis virus (IBV) infection. This study characterized the induction kinetics and anti-IBV functions of these ISGs, and explored the mechanisms underlying their differential induction.

Summary: Wildlife is a reservoir for emerging viruses, and this study identified and characterized 27 families of mammalian viruses from wild and zoo animals in south China. Bats were found to have a high diversity of coronaviruses, picornaviruses, and astroviruses, as well as a potentially new genus of Bornaviridae. The study also discovered potential cross-species transmission of RNA and DNA viruses between wildlife and domestic animals, complicating disease prevention and control. Overall, the study provides important insights into zoonotic risk and the frequency of host-jumping events.

Next-generation diagnostic test for dengue virus detection using an ultrafast plasmonic colorimetric RT-PCR strategy

Summary: The researchers have developed a simple and low-cost plasmonic photothermal (PPT)-reverse transcription-colorimetric polymerase chain reaction (RTcPCR) for molecular diagnosis of dengue virus (DENV) infection. The assay can be completed within 54 minutes and has a detection limit of 1.6 copies/μL of viral nucleic acid. The clinical performance evaluation showed a diagnostic agreement of 97.5% compared to the reference RT-qPCR.
