Luteolin exerted less inhibitory effect on macrophage activation induced by Astragalus polysaccharide than by lipopolysaccharide

As a kind of food supplementary, polysaccharide-rich tonic herbs become more and more popular. However, little is known regarding the interaction between known active molecules with these polysaccharides. This study aims to investigate effects of luteolin on RAW264.7 cells induced by Astragalus polysaccharide RAP in comparison with lipopolysaccharide (LPS). The results showed that luteolin suppressed inflammatory meditors in LPS-induced RAW264.7 cells. While in the RAP-induced RAW264.7 cells, luteolin only decreased the NO production and iNOS gene expression, but only inhibit IL-6 at high concentration, and was ineffective in term of TNF-alpha. The results of western blot analysis suggested that luteolin only inhibited the phosphorylation of NF-kappa B p65 and Akt, but not MAPKs and 1RF3 in RAP-induced RAW264.7 cells. Taken together, lutelion exerted less inhibitory effect on macrophage induced by RAP than by LPS. This study demonstrated the luteolin-containing foods may not strongly interfere the immunoregulation effect of polysaccharide RAP. (C) 2017 Elsevier Ltd. All rights reserved.

Summary: The gut barrier poses a significant research challenge in studying orally bioactive polysaccharides in both in vivo and in vitro studies, particularly in understanding how these polysaccharides interact with related cells in vivo. This study examines the antitumor activity of a hyperbranched heteroglycan RAP from Radix Astragali, showing that its activity is immune-dependent and that it enters Peyer's patches to target dendritic cells and initiate antitumor immune responses. These findings provide insights into a lymphatic route for orally administered RAP to directly interact with immune cells and trigger antitumor immune responses, bridging the gap between in vitro and in vivo studies.

CD30 Regulation of IL-13-STAT6 Pathway in Breast Implant-Associated Anaplastic Large Cell Lymphoma

Summary: This study investigated the unique tumor pathology and growth regulation of Breast implant-associated anaplastic large cell lymphoma (BIA-ALCL) and explored potential targeted therapies. The results showed that CD30 expression is required for IL-13-STAT6 signaling and p38 MAPK activation in BIA-ALCL tumor cells. Inhibitors targeting the IL-13 pathway and p38 MAPK may be effective in treating BIA-ALCL patients.

Summary: In this study, an oligosaccharide sequencing analysis was used to successfully correct the backbone of Dendrobium officinale polysaccharide (DOP) to mannan. This analysis involved producing oligosaccharide fragments through mild acid hydrolysis of DOP followed by p-aminobenzoic ethyl ester (ABEE) derivatization. The isolated and identified oligosaccharides with varied polymerization degrees (DP = 2-10) confirmed that the backbone of DOP is a beta-D-1,4-mannan. This oligosaccharide analysis is more straightforward and reliable compared to conventional methods.

Summary: This study evaluated the effect of cinnamon extract on bowel symptoms in rat models of irritable bowel syndrome with diarrhea (IBS-D). It found that cinnamon extract reduced defecation frequency and visceral hyperalgesia in the rat models, and the efficacy was comparable to a positive drug. Furthermore, cinnamon extract directly inhibited the expression of Tph1 and controlled serotonin synthesis, providing a scientific viewpoint for the traditional use of cinnamon in treating IBS.

Dehydroeburicoic Acid, a Dual Inhibitor against Oxidative Stress in Alcoholic Liver Disease

Summary: This study reports the use of dehydroeburicoic acid (DEA) as a dual inhibitor of the Keap1-Nrf2 protein-protein interaction (PPI) and GSK3 beta, providing potential therapeutic strategies for alcoholic liver disease (ALD). DEA disrupts the Keap1-Nrf2 PPI and restores Nrf2 activity, leading to the activation of antioxidant genes. It also alleviates ethanol-induced mitochondrial dysfunction and exhibits minimal toxicity in the ALD cell model.

Affinity-Based Luminescent Iridium(III) Complexes for the Detection of Disease-Related Proteins

This review summarizes the current advances and future perspectives of affinity-based luminescent iridium(III) complexes for detecting disease-related proteins.

Summary: This study revealed the critical role of the interleukin-1R (IL-1R) inflammatory pathway in pC-ALCL and its potential as a therapeutic target. The IL-1R pathway was found to promote JAK-STAT3 signaling activation and enhance the sensitivity of pC-ALCL cells to JAK inhibitors. The dual inhibitor pacritinib showed strong activities against pC ALK(-) ALCL in vitro and in vivo.

An oligosaccharide marker for rapid authentication of edible bird?s nest

Summary: In this study, a specific oligosaccharide marker for rapid authentication of edible bird's nest (EBN) was identified by comparing the saccharide profiles of EBN and adulterants. This marker, named BNM001, was characterized as Neu5Ac (2-3) Gal by MS and NMR spectra. The ABEE labeling of this marker improved the accuracy and sensitivity of LC-qTOF-MS quantitative analysis. The relative marker content was found to be associated with the quality of EBN products.
