Background: Survival differences across Europe for patients with cancers of breast, uterus, cervix, ovary, vagina and vulva have been documented by previous EUROCARE studies. In the present EUROCARE-5 study we update survival estimates and investigate changes in country-specific and over time survival, discussing their relationship with incidence and mortality dynamics for cancers for which organised screening programs are ongoing. Methods: We analysed cases archived in over 80 population-based cancer registries in 29 countries grouped into five European regions. We used the cohort approach to estimate 5-year relative survival (RS) for adult (P15 years) women diagnosed 2000-2007, by age, country and region; and the period approach to estimate time trends (1999-2007) in RS for breast and cervical cancers. Results: In 2000-2007, 5-year RS was 57% overall, 82% for women diagnosed with breast, 76% with corpus uteri, 62% with cervical, 38% with ovarian, 40% with vaginal and 62% with vulvar cancer. Survival was low for patients resident in Eastern Europe (34% ovary-74% breast) and Ireland and the United Kingdom [Ireland/UK] (31-79%) and high for those resident in Northern Europe (41-85%) except Denmark. Survival decreased with advancing age: markedly for women with ovarian (71% 15-44 years; 20% P75 years) and breast (86%; 72%) cancers. Survival for patients with breast and cervical cancers increased from 1999-2001 to 2005-2007, remarkably for those resident in countries with initially low survival. Conclusions: Despite increases over time, survival for women's cancers remained poor in Eastern Europe, likely due to advanced stage at diagnosis and/or suboptimum access to adequate care. Low survival for women living in Ireland/UK and Denmark could indicate late detection, possibly related also to referral delay. Poor survival for ovarian cancer across the continent and over time suggests the need for a major research effort to improve prognosis for this common cancer. (C) 2015 Elsevier Ltd. Summary: This study provides a comprehensive examination of worldwide variation and trends in survival from brain tumors in adults by analyzing individual data from 2000 to 2014. The results show significant regional differences and temporal changes in survival rates, especially for the most common sub-types, with remarkable improvements since 2005 due to the uptake of chemoradiation. However, there are still some countries that lag behind in terms of survival rates.

Summary: Plasmapheresis donation is generally safe, but its long-term effects remain unclear. The amount of anticoagulant used in the process varies, and it is influenced by factors such as hematocrit, total blood processed, plasma collected, and donor characteristics. This study found that the unit weight setting and sex have an impact on anticoagulant distribution and post-donation citrate metabolism.

Vitamin D Status, Cdx2 Genotype, and Colorectal Cancer Survival: Population-Based Patient Cohort

Tafirenyika Gwenzi, Petra Schrotz-King, Ben Schoettker, Michael Hoffmeister, Hermann Brenner

Summary: According to recent evidence, the prognostic value of Vitamin D (VitD) status for colorectal cancer (CRC) patients might be confined to patients with the GG genotype of Cdx2, a functional polymorphism of the VitD receptor gene. In this study, the researchers aimed to validate these findings in a cohort of CRC patients by measuring serum VitD concentration and performing Cdx2 genotyping. The results showed that VitD deficiency is associated with poorer survival, particularly in patients with the GG genotype of Cdx2.

Estimating complete cancer prevalence in Europe: validity of alternative vs standard completeness indexes

Elena Demuru, Silvia Rossi, Leonardo Ventura, Luigino Dal Maso, Stefano Guzzinati, Alexander Katalinic, Sebastien Lamy, Valerie Jooste, Corrado Di Benedetto, Roberta De Angelis

Summary: Comparable indicators on complete cancer prevalence are needed in Europe. Two alternative approaches, empirical indices derived from European registries and pre-calculated indices derived from US-SEER cancer registries, were compared to standard estimates using data from 62 registries in 27 countries. Empirical estimates differ little from standard estimates for elderly-diagnosed tumors, but may underestimate complete prevalence for early-onset cancers. SEER estimates are comparable to standard ones for most cancers, even derived from short time series, but longer observations are needed for certain cancers with remarkable differences in incidence and prognosis between European and US populations.

Summary: This study aimed to assess whether the effect of high temperature on mortality differed in COVID-19 survivors and naive individuals. Data from summer mortality and COVID-19 surveillances were used. The results showed a 3.8% excess risk in summer 2022 compared to 2015-2019, with a 20% increase in the last fortnight of July, the period with the highest temperature. The increase in mortality rates during this period was higher among naive individuals compared to COVID-19 survivors. Time series analysis confirmed the association between temperatures and mortality in naive individuals but not in COVID-19 survivors.

Summary: Cancer registration in Germany has a long tradition and in 2013, new legislation mandated the implementation of additional clinical cancer registration. The aim was to measure and improve the quality of cancer care. The current status of extended cancer registration in Germany shows a slight decrease in the age-standardized incidence of cancer over the last decade. The five-year relative survival rate for all cancers is 67% for women and 63% for men.

Potential of Fecal Carcinoembryonic Antigen for Noninvasive Detection of Colorectal Cancer: A Systematic Review

Xianzhe Li, Lara Stassen, Petra Schrotz-King, Zitong Zhao, Rafael Cardoso, Janhavi R. Raut, Megha Bhardwaj, Hermann Brenner

Summary: This systematic review evaluated fecal carcinoembryonic antigen (FCEA) as a biomarker for the noninvasive diagnosis and diagnosis of colorectal cancer (CRC). The study found significant differences in FCEA levels between CRC and control groups, and the diagnostic performance of FCEA was superior to that of serum carcinoembryonic antigen (SCEA), suggesting its potential role as a novel and easily measurable biomarker for the diagnosis of CRC. However, more comprehensive evaluation in screening settings is needed due to limited evidence from clinical studies.

The results show that ATAX does not increase the risk of MACE and there is no difference in survival between patients who received TAX and ATAX.

Immunotherapy response in microsatellite stable metastatic colorectal cancer is influenced by site of metastases

Marwan Fakih, Chongkai Wang, Jaideep Sandhu, Jian Ye, Colt Egelston, Xiaochen Li

Summary: This study explores the impact of metastatic sites on treatment outcomes for chemotherapy-refractory colorectal cancer patients. It found that patients with liver or peritoneal metastases had poor treatment outcomes, while those with lung-only metastases showed significant response. The presence of concurrent lymph node or other extrahepatic metastatic disease diminished treatment response in patients with lung metastases. Future checkpoint inhibitor trials should stratify patients based on metastatic locations.

Summary: The study reveals that nullpeptides can serve as biomarkers for cancer detection and treatment, particularly in highly recurrent cancer patients. These nullpeptides primarily occur in highly expressed genes, particularly in specific loci of oncogenes and tumor suppressors. Recurrent nullpeptides are more likely to be found in neoantigens, which play a significant role in immunotherapy.
