Antibodies (Abs) produced by immunoglobulin (IG) genes are the most diverse proteins expressed in humans. While part of this diversity is generated by recombination during B-cell development and mutations during affinity maturation, the germ-line IG loci are also diverse across human populations and ethnicities. Recently, proof-of-concept studies have demonstrated genotype phenotype correlations between specific IG germ-line variants and the quality of Ab responses during vaccination and disease. However, the functional consequences of IG genetic variation in Ab function and immunological outcomes remain underexplored. In this opinion article, we outline interconnections between IG genomic diversity and Ab-expressed repertoires and structure. We further propose a strategy for integrating IG genotyping with functional Ab profiling data as a means to better predict and optimize humoral responses in genetically diverse human populations, with immediate implications for personalized medicine.

Summary: This study reveals a novel role for CD11b in regulating the healthy humoral response and autoimmune diseases, with CD11b deficiency leading to an enhanced specific humoral response in mice. Vaccine genetics of IGHV1-2 VRC01-class broadly neutralizing antibody precursor naive human B cells

Summary: The study demonstrates that a successful HIV vaccine must activate naive precursor B cells encoding broadly neutralizing antibodies, and the use of germline-targeting immunogens and high-throughput single-cell BCR sequencing can efficiently screen large numbers of BCR sequences to elucidate precursor frequencies. Furthermore, analysis of IGHV1-2 allelic usage across different cohorts reveals common alleles previously thought incompatible with VRC01-class responses, and germline variation within IGHV1-2 associates with gene usage frequencies in the naive BCR repertoire.

Age-associated B cells are heterogeneous and dynamic drivers of autoimmunity in mice

Summary: Age-associated B cells (ABCs) are a type of memory B cell expressing the transcription factor T-bet, formed under inflammatory conditions. In systemic lupus erythematosus (SLE), ABC frequency is correlated with disease and they are considered the source of autoantibody-secreting cells. The heterogeneity of ABCs and their relationship to other B cell subsets such as plasmablasts is unclear. The study shows that ABCs are dynamic and can be plasmablast precursors or undergo cycles of reactivation. Deletion of CD11c-expressing B cells reveals a direct role for ABC-like B cells in lupus pathogenesis.

Summary: In adaptive immune receptor repertoire analysis, determining the germline variable (V) allele associated with each T- and B-cell receptor sequence is challenging. In this study, we propose an alternative naming scheme for V alleles and a novel method to infer individual genotypes. We validate the effectiveness of our approach and implement it in a freely available R package (PIgLET).
