BiHC, a T-Cell-Engaging Bispecific Recombinant Antibody, Has Potent Cytotoxic Activity Against Her2 Tumor Cells

Among different cancer immunotherapy approaches, bispecific antibodies (BsAbs) are of great interest due to their ability to recruit immune cells to kill tumor cells directly. Various BsAbs against Her2 tumor cells have been proposed with potent cytotoxic activities. However, most of these formats require extensive processing to obtain heterodimeric bispecific antibodies. In this study, we describe a bispecific antibody, BiHC (bispecific Her2-CD3 antibody), constructed with a single-domain anti-Her2 and a single-chain Fv (variable fragment) of anti-CD3 in an IgG-like format. In contrast to most IgG-like BsAbs, the two arms in BiHC have different molecular weights, making it easier to separate hetero-or homodimers. BiHC can be expressed in Escherichia coli and purified via Protein A affinity chromatography. The purified BiHC can recruit T cells and induce specific cytotoxicity of Her2-expressing tumor cells in vitro. The BiHC can also efficiently inhibit the tumor growth in vivo. Thus, BiHC is a promising candidate for the treatment of Her2-positive cancers.

A novel isoform of hydroxyacyl-CoA dehydrogenase inhibits cell proliferation

Summary: In this study, we identified two alternative translation start codons in Hadh mRNA using QTI-seq. We found that translation from the upstream start codon encodes the mitochondrial isoform of HADH, while translation from the downstream start codon produces a short isoform (HADH-S) with predominant nuclear localization. Additionally, overexpression of HADH-S was shown to inhibit cell proliferation.

Improvement of mechanical properties of hot extruded and aged Mg-Zn-Mn-Sn alloy through Dy addition

Summary: The effects of different Dy additions on the microstructure and mechanical properties of Mg-6Zn-1Mn-3Sn (wt%, ZMT613) alloys were investigated. The study found that Dy addition resulted in significant grain refinement and the formation of new MgSnDy phases. A small amount of Dy (≤0.8 wt%) hindered the proportion of dynamic recrystallized grains during the hot-extrusion process and enhanced the basal texture of the alloy. MgSnDy high-temperature phase particles inhibited the recovery process and promoted the precipitation of the second phase. The ZMT613-0.2Dy alloy showed optimum mechanical properties after aging, with yield strength, ultimate tensile strength, and elongation reaching 336 MPa, 399 MPa, and 4.4%, respectively.

The formation mechanism of Al-RE tubular phase in AZ80-Ce magnesium alloy by La doping

Summary: This paper investigates the effect of La addition on the Al-RE phase in Mg-Al-Zn-Ce magnesium alloy. The results show that adding La can change the morphology of the Al11RE3 phase and a transformation mechanism is proposed. La replaces part of Ce in the Al11Ce3 phase, resulting in the formation of Al11RE3 phase. In the Al11RE3 phase, La doping leads to lattice distortion and accelerates the growth rate in a specific direction, forming a tubular structure.

Two-stage screened selection designs for randomized phase II trials with time-to-event endpoints

A high-strength Mg-8Zn-1Mn-3Sn-1.2Gd alloy with fine MgSnGd particles by Dy modification

Summary: In this work, an Mg-8Zn-1Mn-3Sn-1.2Gd-0.2Dy alloy with excellent combination of tensile strength and ductility is successfully prepared. The formation mechanism of Dy-modified MgSnGd phase and its effect on the microstructure and mechanical properties of the alloy are systematically investigated. The addition of Dy promotes dynamic recrystallization and inhibits the growth of MgSnGd phase, resulting in the alloy exhibiting high strength and ductility.

A semi-mechanistic dose-finding design in oncology using pharmacokinetic/pharmacodynamic modeling

Summary: This paper proposes a new framework for modeling the dose-response relationship in oncology phase I dose-finding designs by incorporating pharmacokinetic data and the hypothesized mechanisms of drug effects. The resulting design improves upon common designs and considers the impact of dose, schedule, and method of administration on toxicity. The performance of this design is comparable to other designs when there is significant departure from the underlying drug effect mechanism.

Pediatric Craniopharyngioma: The Effect of Visual Deficits and Hormone Deficiencies on Long-Term Cognitive Outcomes After Conformal Photon Radiation Therapy

Summary: This study investigated the cognitive outcomes of pediatric patients with craniopharyngioma after conformal photon radiation therapy and the effects of visual deficits and hormone deficiencies. The results showed that younger age and higher doses to the temporal lobes and hippocampi were associated with lower intellectual functioning. Black race and cerebrospinal fluid shunting were linked to lower baseline intelligence quotient. Patients treated for growth hormone deficiency within 12 months of radiation therapy had better intelligence and attention outcomes. It is recommended to improve treatment selection, minimize damage to normal tissues, and manage endocrinopathy in pediatric craniopharyngioma patients.

Recent Use of Pediatric Extrapolation in Pediatric Drug Development in US

Summary: The regulatory standards of the FDA require substantial evidence of effectiveness for drugs developed in both adults and children, but extrapolation may still be acceptable. This study reviewed pediatric labeling changes published by the FDA between 1/1/2015 and 7/31/2021, and found that 63.8% of the drugs identified used extrapolation. Extrapolation was most commonly used in neuroscience and infectious disease, and its use increased the rates of new and expanded pediatric indications.

A Bayesian two-stage group sequential scheme for ordinal endpoints

However, design methods for ordinal endpoints are limited, and a dichotomized approach is often used for simplicity. This study introduces a Bayesian group sequential scheme to assess ordinal endpoints, considering various scenarios and providing desirable performance through extensive simulations. The R package BayesOrdDesign is publicly available.

Diagnostic plasma small extracellular vesicles miRNA signatures for pancreatic cancer using machine learning methods

Ceramide metabolism-related prognostic signature and immunosuppressive function of ST3GAL1 in osteosarcoma
