The objective of this study was to develop and validate a fast, sensitive and simple liquid chromatography-electrospray ionization-tandem mass spectrometry (LC-ESI-MS/MS) method for the screening of six classes of antibiotics (aminoglycosides, beta-lactams, macrolides, quinolones, sulfonamides and tetracyclines) in fish. Samples were extracted with trichloroacetic acid. LC separation was achieved on a Zorbax Eclipse XDB C18 column and gradient elution using 0.1% heptafluorobutyric acid in water and acetonitrile as mobile phase. Analysis was carried out in multiple reaction monitoring mode via electrospray interface operated in the positive ionization mode, with sulfaphenazole as internal standard. The method was suitable for routine screening purposes of 40 antibiotics, according to EC Guidelines for the Validation of Screening Methods for Residues of Veterinary Medicines, taking into consideration threshold value, cut-off factor, detection capability, limit of detection, sensitivity and specificity. Real fish samples (n=193) from aquaculture were analyzed and 15% were positive for enrofloxacin (quinolone), one of them at a higher concentration than the level of interest (50 mu g kg(-1)), suggesting possible contamination or illegal use of that antibiotic.

Germinated sorghum (Sorghum bicolor L.) and seedlings show expressive contents of putrescine

Summary: This study quantified the levels of free bioactive amines during germination of tannin and tannin-free sorghum seeds and seedlings. It was found that tannin-free sorghum seeds had higher levels of spermidine, spermine, putrescine, and total amines compared to tannin sorghum. Throughout germination, putrescine was prevalent followed by spermidine. Spermine was only detected in the 7th germination day in tannin sorghum. The findings suggest that by selecting different genotypes, the polyamine contents in germinated sorghum can be modulated for specific food applications.

Thermoresponsive in-situ gel containing hyaluronic acid and indomethacin for the treatment of corneal chemical burn

Summary: Ocular chemical burns can be effectively treated with a poloxamer-based thermoresponsive in-situ gelling system containing hyaluronic acid and indomethacin. The optimized system (F2) has a gelling temperature of 34.2 degrees C and shows improved wound healing in the ocular surface.

Varietal Influence on the Formation of Bioactive Amines during the Processing of Fermented Cocoa with Different Pulp Contents

Summary: The presence and levels of bioactive amines during the processing of cocoa were investigated in this study. The fermentation time, pulp content, and cocoa variety were found to influence the content of bioactive amines. Cadaverine was detected during drying in all cocoa varieties, reaching levels of up to 17.96 mg/kg dwb. Most bioactive amines decreased during roasting, except for phenylethylamine, which increased.

Tocotrienols-enriched Self-nanoemulsifying Drug Delivery System Enhances the Antileukemic Activity of All-trans Retinoic Acid but not Electrocardiogram Alterations Evoked by Its Combination with Arsenic Trioxide

Summary: All-trans retinoic acid and arsenic trioxide are effective treatments for acute promyelocytic leukemia, but they have certain drawbacks such as patient relapse and cardiotoxicity. In this study, a self-nanoemulsifying drug delivery system enriched with tocotrienols was developed to enhance the cytotoxicity of all-trans retinoic acid and mitigate the cardiotoxic effects of arsenic trioxide. The system showed promising results in improving the antileukemic properties of all-trans retinoic acid, but it also influenced the electrocardiogram changes caused by arsenic trioxide administration.

Thermal energy and tableting effects in benznidazole product: the impacts of industrial processing

Summary: This study evaluated the thermal energy and tableting effects of benznidazole (BZN), excipients, and tablets using techniques such as infrared spectroscopy, X-ray diffraction, and thermal analysis. The results revealed the dehydration of talc and alpha-lactose monohydrate and the conversion of beta-lactose to stable alpha-lactose during tableting. The findings highlight the importance of considering thermal energy and tableting effects in the manufacturing of BZN.

Fatty acid profile and rheological behavior of annatto seed oil (Bixa orellana), cupuassu seed fat (Theobroma grandiflorum), and their blends

Summary: This study evaluated the fatty acid composition, nutritional quality, FTIR spectra, and rheological behavior of blends of annatto seed oil (ASO) and cupuassu seed fat (CSF) at different ratios (30:70, 50:50, and 70:30 % w/w). The results showed that ASO increased the content of polyunsaturated fatty acids in the blends, while CSF conferred higher contents of monounsaturated fatty acids. The blends exhibited low atherogenicity and thrombogenicity indices, indicating their nutritional advantages. The Newtonian fluid behavior and FTIR results suggested that mixing ASO and CSF at different proportions did not affect the functional groups. ASO showed higher activation energy value, indicating its viscosity was more affected by temperature changes. The Newtonian model proved suitable for describing the behavior of the samples.

Microextraction by packed sorbent and high-performance liquid chromatography for determination of benznidazole in human plasma

Summary: This study aimed to develop and validate a MEPS coupled to high performance liquid chromatography method for the analysis of benznidazole in human plasma. The method was proved to be selective, precise, accurate, robust and linear in the range from 0.5 to 6.0 μg mL-1, and it was applied successfully to assess this drug in plasma samples.

Intravitreal acetazolamide implant for pseudophakic cystoid macular edema

Summary: This article reports a case of pseudophakic cystoid macular edema (PCME) in which conventional treatment was ineffective but improved after intravitreal acetazolamide implant injection. Complete resolution of symptoms was observed 4 weeks after the injection. Further investigation is needed to confirm these findings.

Sweeteners in Brazilian processed foods and beverages: prevalence, profile and concomitant addition of sugars and nutritional claims

Summary: Brazil is facing a growing problem of overweight and obesity, and various measures have been taken to address this issue, such as reducing sugar intake and implementing new packaging labeling. As a result, the use of low or non-caloric sweeteners (LNCS) has increased. This study examined the presence of LNCS in six food groups in Brazil, and found that 17.7% of the included products contained at least one LNCS, with an average of 2.21 LNCS per sweetened product. Non-alcoholic beverages and dairy products had the highest occurrence of LNCS.

The occurrence of mycotoxins in dry fermented sausage is described worldwide, but little information is available for Brazilian products. This study investigated the occurrence of aflatoxins (B1, B2, G1, and G2) and ochratoxin A (OTA) in dry fermented sausage using a validated dilute and shoot method.

Summary: In this study, a fluorescent biosensing system for high sensitivity detection of MT was established using a new Ag+-stabilized DNA triplex probe (Ag+-SDTP) combined with catalytic hairpin assembly (CHA) and CRISPR/Cas12a signal enhancements. The sensing method is able to selectively detect MT from other non-specific molecules and can achieve low level detection of MT in diluted human serums, demonstrating its potential for monitoring disease-specific MT biomarker at trace levels.

A beta 1-42 and A beta 1-40 have been identified as relevant biomarkers for AD diagnosis, and SurfaceAssisted Laser Desorption/Ionization Mass Spectrometry (SALDI-MS) was used to monitor the A beta 1-42/A beta 1-40 ratio without prior sample treatment.

CRISPR-empowered electrochemical biosensor for target amplification-free and sensitive detection of miRNA

Summary: We developed a CRISPR-empowered electrochemical biosensor, named PER-E-CRISPR, for amplification-free and sensitive detection of miR-21. The biosensor combines the advantages of CRISPR/Cas13a and primer exchange reaction (PER) for target amplification. Under optimized conditions, the PER-E-CRISPR assay exhibits a linear range from 10-13 to 10-7 M and a limit of detection of 30.2 fM. The biosensor shows excellent performance in actual plasma, indicating promising prospects for miRNA detection in molecular diagnosis.

Application and evaluation of molecular docking for aptamer and small molecular interaction- A case study with tetracycline antibiotics

Summary: Molecular docking (MD) analysis is widely used for studying the interaction between aptamers and small molecules. This study established steady-state aptamers of tertiary structures (SATS) using specific aptamers of tetracycline antibiotics as docking models. Molecular docking results revealed multiple binding sites in the SATS of aptamers, with significant variations in binding free energy (BFE) and docking score (DS). The study also proposed a method for MD analysis based on SATS, providing insights into binding mode and predicting binding sites.

The construction of highly selective surface molecularly imprinted polymers based on Cu(II) coordination for the detection of bisphenol A

Summary: In this study, a highly selective molecularly imprinted polymer Cu-MIPs@CS was designed and synthesized for bisphenol A (BPA). Cu-MIPs@CS showed significantly enhanced imprinting factor and selective factor towards BPA compared to MIPs@CS. It also exhibited superior discrimination ability between BPA and its structural analogue. Additionally, Cu-MIPs@CS were successfully applied as a solid phase extraction adsorbent for the determination of BPA in drinking water samples.

A simple joint detection platform for high-throughput single-cell heterogeneity screening
