Summary: Recent studies have identified a previously unknown function for the lncRNA GAPLINC, which acts as a negative regulator of inflammation in the context of innate immunity, macrophages, and LPS-induced endotoxic shock. Transcriptome analysis in human and mouse macrophages showed that GAPLINC is highly expressed following macrophage differentiation, but rapidly down-regulated upon inflammatory activation. Depletion of GAPLINC leads to enhanced expression of inflammatory genes, while overexpression suppresses this response, ultimately modulating endotoxic shock and multiorgan failure.

Summary: This study aims to establish the gender specificity pattern from immune cell infiltration and four kinds of cell-death perspectives to improve ischemic stroke (IS) diagnosis and therapy. The results showed significant changes in immune cell infiltration and the identification of gender-specific cell-death related genes. Machine learning techniques further revealed diagnostic models and biomarkers. These findings contribute to a better understanding of immune cell infiltration and cell death mechanisms, providing valuable therapeutic targets for IS patients of different genders.

MPLA induced HLA family genes more strongly than LPS in both populations, suggesting divergent signaling pathways downstream of TLR-4 and better stratification potential within the septic population.

Summary: In this study, the protective role and mechanisms of IRG1/itaconate on LPS-induced multi-organ injury were explored. It was found that IRG1 deficiency aggravated the injury while 4-Octyl itaconate significantly ameliorated the acute lung, liver, and kidney injury induced by LPS. Furthermore, IRG1/4-OI protected against the injury by inhibiting inflammation response and GSDMD-mediated pyroptosis.
