Oral supplementation of inorganic pyrophosphate in pseudoxanthoma elasticum

Eszter Kozak, Krisztina Fulop, Natalia Tokesi, Nidhi Rao, Qiaoli Li, Sharon F. Terry, Jouni Uitto, Xiaoming Zhang, Cyrus Becker, Andras Varadi, Viola Pomozi

Summary: PXE is a rare hereditary multisystem disorder affecting elastic fibers in the skin, eyes, and cardiovascular system. Research has shown that orally administered pyrophosphate compounds can inhibit ectopic calcification, providing a crucial foundation for potential clinical trials involving oral PPi in the future.

Genetic heterogeneity of heritable ectopic mineralization disorders in a large international cohort

Summary: This study reports the genetic findings from an international cohort of patients with heritable ectopic mineralization. Multiple gene variants were identified, including 159 distinct variants in the ABCC6 gene. The functional assessment of these variants highlighted the importance of determining pathogenicity through appropriate experimental systems.

ENPP1 variants in patients with GACI and PXE expand the clinical and genetic heterogeneity of heritable disorders of ectopic calcification

Douglas Ralph, Yvonne Nitschke, Michael A. Levine, Matthew Caffet, Tamara Wurst, Amir Hossein Saeidian, Leila Youssefian, Hassan Vahidnezhad, Sharon F. Terry, Frank Rutsch, Jouni Uitto, Qiaoli Li

Summary: This study reveals that variants in the ENPP1 gene can cause both GACI and classic PXE, increasing the clinical and genetic diversity of hereditary ectopic calcification disorders. Additionally, the correlation between plasma PPi levels and the severity of vascular calcification is found to be limited in this study.

The pathogenic c.1171A>G (p.Arg391Gly) and c.2359G>A (p.Val787Ile) ABCC6 variants display incomplete penetrance causing pseudoxanthoma elasticum in a subset of individuals

Flora Szeri, Agnes Miko, Nastassia Navasiolava, Ambrus Kaposi, Shana Verschuere, Beatrix Molnar, Qiaoli Li, Sharon F. Terry, Federica Boraldi, Jouni Uitto, Koen van de Wetering, Ludovic Martin, Daniela Quaglino, Olivier M. Vanakker, Kalman Tory, Tamas Aranyi

Summary: Incomplete penetrance is a source of heterogeneity in pseudoxanthoma elasticum. Two incomplete penetrant pathogenic variants were identified, but they are only deleterious when a yet unknown interacting partner of ABCC6 is mutated simultaneously.

Summary: This study aimed to investigate the effects of different mindfulness interventions on overall well-being and whether baseline characteristics would have an impact on the treatment outcomes. The results showed that the standard 8-session MBCT program improved well-being, but it was not superior to the brief 3-session mindfulness intervention.

A new enzymatic assay to quantify inorganic pyrophosphate in plasma

Stefan Lundkvist, Fatemeh Niaziorimi, Flora Szeri, Matthew Caffet, Sharon F. Terry, Gunnar Johansson, Robert S. Jansen, Koen van de Wetering

Summary: Inorganic pyrophosphate (PPi) is a crucial regulator of extracellular mineralization, but there is currently no reliable assay for its quantification. In this study, we report a new and robust assay that converts PPi into ATP and quantifies it using firefly luciferase-based bioluminescence. The assay shows high sensitivity, precision, and accuracy, and outperforms currently available assays.
