Focal adhesion kinase (FAK) is a nonreceptor tyrosine kinase involved in development and human disease, including cancer. It is currently thought that the four-point one, ezrin, radixin, moesin (FERM)- kinase domain linker, which contains autophosphorylation site tyrosine (Y) 397, is not required for in vivo FAK function until late midgestation. Here, we directly tested this hypothesis by generating mice with FAK Y397-to-phenylalanine (F) mutations in the germline. We found that Y397F embryos exhibited reduced mesodermal fibronectin (FN) and osteopontin expression and died during mesoderm development akin to FAK kinase-dead mice. We identified myosin-1E (MYO1E), an actin-dependent molecular motor, to interact directly with the FAK FERM-kinase linker and induce FAK kinase activity and Y397 phosphorylation. Active FAK in turn accumulated in the nucleus where it led to the expression of osteopontin and other FN-typematrix in both mouse embryonic fibroblasts and human melanoma. Our data support a model in which FAK Y397 autophosphorylation is required for FAK function in vivo and is positively regulated by MYO1E.

The Emerging Terbinafine-Resistant Trichophyton Epidemic: What Is the Role of Antifungal Susceptibility Testing?

Julia J. Shen, Maiken C. Arendrup, Shyam Verma, Ditte Marie L. Saunte

Summary: Dermatophytosis is commonly encountered in dermatological clinics, with Trichophyton rubrum, T. mentagrophytes, and T. interdigitale as the main causative agents. Terbinafine therapy is effective but increasing resistance and mutations in the SQLE gene have been documented. Antifungal susceptibility testing is not routinely performed for dermatophytes, leading to difficulties in interpreting MICs.

Incidence and remission rates of self-reported hidradenitis suppurativa - A prospective cohort study conducted in Danish blood donors

Summary: The incidence and remission rates of self-reported HS are high, with factors such as gender, smoking, and BMI affecting both the development and recovery of HS. Females with BMI above 25, active smokers, and individuals with symptoms in multiple areas are more likely to develop self-reported HS, while weight loss and smoking cessation can significantly impact the remission rate.

Increasing Terbinafine Resistance in Danish Trichophyton Isolates 2019-2020

Summary: Terbinafine resistance in Trichophyton species is increasing in Denmark, with a high prevalence of non-wildtype isolates and known/novel SQLE mutations. T. rubrum is the most common species, but T. mentagrophytes/interdigitale SC isolates also contribute to resistance. Population-based studies and susceptibility testing are important for understanding and addressing this emerging problem.

Summary: This study validates the novel IHS4-55 outcome measure by showing its significant association with reduction in inflammatory lesion counts and achievement of MCIDs for DLQI, NRS Pain, and NRS Pruritus in an antibiotic-treated cohort. These findings support the use of IHS4-55 as a novel primary outcome measure in clinical trials.

Summary: Malassezia folliculitis occurs when the benign colonization of hair follicles by the yeast Malassezia becomes symptomatic, leading to itchy papules and pustules. However, diagnosing and treating this condition varies among dermatologists and countries. The European Academy of Dermatology and Venereology Mycology Task Force has developed recommendations to guide the diagnosis and management of Malassezia folliculitis, including diagnostic methods, required positive findings before treatment, and specific treatment algorithms for different patient groups. Overall, this study provides a clinical strategy for diagnosing and managing Malassezia folliculitis.
