Summary: Melanoma is an immune sensitive disease, and immune check point blockade has shown activity. However, TIL therapy after ICB failure has shown promising efficacy, indicating the potential of cellular therapies. To overcome limitations of TIL treatment, a controlled adoptive cell therapy approach using synthetic agonistic receptors and bispecific antibodies targeting melanoma-associated antigens is proposed.

Summary: The study identified Cbl-b as a potential target for overcoming exhausted CAR T-cell function in solid tumors. Inhibition of Cbl-b restored effector function of exhausted T cells. Depletion of Cbl-b enhanced CAR T-cell efficacy in reducing tumor growth, decreasing exhausted T cells, and increasing effector cytokine expression.

Summary: This study found that depleting naive T cells before extracting antigen-specific T cells from patients can enhance their expansion, antigen specificity, and therapeutic potency. By using CD45RA-depleted PBMCs, EBV-specific T cells were successfully generated and applied in clinical trials for lymphoma and viral infections.

Summary: The interaction between melanoma cells and T cells can enhance the chemotaxis of new T cells. ADAR1-p150 expression is correlated with immune infiltration and can be induced by immunotherapy. ADAR1 regulates T cell migration through the secretion of chemokines.

Summary: The study introduces a lysosome-targeting nanoparticle developed from a mineralized metal-organic framework coupled with a lysosome-targeting aptamer to enhance the antitumor effect of T cells. The MOF synthesized from Zn2+ and dimethylimidazole has good protein encapsulation and acid sensitivity, making it an ideal lysosomal delivery vector. The use of calcium carbonate to induce MOF mineralization improves stability and provides synergistic effects for encapsulating therapeutic proteins.

Summary: With the continuous advancements in immunotherapy and precision medicine, adoptive cell therapy (ACT) has emerged as a new treatment approach in oncology. Chimeric antigen receptor (CAR) T cells, genetically modified lymphocytes, have shown promising results in targeting and killing cancer cells. Commercialization of CAR T cell therapy has paved the way for future bright developments.

Summary: Resident memory CD8 T cells, characterized by their ability to reside in peripheral tissues, play a crucial role in adaptive sentinel activity and amplifying immune responses. The study focused on the chemotactic mechanisms governing the localization, retention, and residency of memory CD8 T cells in the ovarian tumor microenvironment. Results indicated that CXCR6 is a key marker for resident memory tumor-specific CD8 T cells, with its deficiency resulting in reduced retention in tumor tissues and poor control of ovarian cancer. Further research is needed to explore the potential of utilizing CXCR6 to enhance resident memory responses in cancer.

Summary: This study reveals the potential of targeting TNFR2 to enhance anti-tumor responses and treat relapse of blood malignancies. The researchers also found that TNFR2 is over-expressed in Tregs from healthy donors and patients with leukemia relapse or GVHD. These findings provide new perspectives for the development of immunotherapies.

Summary: CAR T cells are a type of immunotherapy that has revolutionised the treatment of haematological malignancies by genetically modifying a patient's own cells to target and kill cancer cells. However, obstacles such as the individual production of CAR T cells result in variability in the product, longer wait times for treatment, and higher costs. Novel approaches utilizing allogeneic cells, known as off the shelf CAR T cells, have emerged to overcome these challenges.

Summary: This study found that enforced expression of the high-affinity glucose transporter GLUT3 can improve metabolic fitness of tumor-directed T cells. GLUT3-OT1 T cells showed better tumor control and significantly improved survival in a melanoma model, with some treated mice being cured and protected from re-challenge, indicating long-term T cell persistence and memory formation.
