Improving on Nature: The Role of Nanomedicine in the Development of Clinical Natural Drugs

Natural products have been used as a major source of drugs for millennia, and about half of the pharmaceuticals in use today are derived from natural products. However, their efficacy can be limited because of their low hydrophilicity and intrinsic dissolution rate(s), or physical/chemical instability. In addition, they can present scarce absorption, poor pharmacokinetics and bioavailability, scarce biodistribution, first-pass metabolism, trivial penetration and accumulation in the organs of the body, or low targeting efficacy. Novel nanoformulations based on drug delivery systems, namely nanoparticles, micelles, and vesicles, offer significant promise in overcoming these limitations. Nowadays, nanomedicine is crucial in developing appropriate therapeutic treatments of essential drugs, specifically antitumor and antiparasistic agents (i.e., Taxol, vincristine, camptothecin, doxorubicin, artemisinin) and other emerging molecules with pleiotropic functions (i.e., resveratrol, curcumin, salvianolic acid B, honokiol). Additionally, the number of nanoformulations developed with flavonoids, in particular rutin, quercetin, silymarin, and green tea catechins, is constantly increasing, and a significant number of publications have appeared in the last decade pertaining to nanoformulations based on extracts and essential oils. Most of these studies report very promising nanoformulations with sustained release and improved bioavailability at much lower doses than conventional preparations, and in many cases, also a better safety profile.

Nanostructured Lipid Carriers Can Enhance Oral Absorption of Khellin, a Natural Pleiotropic Molecule

Summary: A novel formulation based on NLCs was developed to enhance the solubility and intestinal absorption of khellin. This versatile formulation has the potential to improve the bioavailability and therapeutic effects of khellin.

Summary: Two liposomal formulations, one of which is coated with hyaluronic acid, have been developed to enhance the solubility of Thymoquinone (TQ) and increase its absorption at the cellular level, especially in the nucleus. These formulations not only decrease TQ toxicity observed at high doses in cells, but also result in a gradual and prolonged release of TQ in both coated and uncoated liposomes.

Development and Evaluation of Khellin-loaded Microemulgel for Dermatological Applications

Summary: Microemulsions are versatile drug delivery systems that can be administered regardless of route. In this study, a microemulsion was formulated using glycerol (2.25%), Labrasol (20.25%), vitamin E acetate (2.50%), and water (75.00%). The microemulsion showed stable globules with a size of about 17 nm and had high potential for skin care applications due to its dermatological properties.

Escinosomes: Safe and Successful Nanovesicles to Deliver Andrographolide by a Subcutaneous Route in a Mice Model of Oxaliplatin-Induced Neuropathy

Summary: This study successfully addressed the low aqueous solubility and limited bioavailability of andrographolide (AG) by encapsulating it in escinosomes, nanovesicles made of escin and phosphatidylcholine. AG-loaded escinosomes exhibited favorable properties and achieved sustained release of AG. Moreover, these nanovesicles effectively alleviated thermal allodynia in a rat model of chemotherapy-induced neuropathy.

Summary: The study formulated oleanolic acid (OA) into two different microemulsions (ME-1 and ME-2) to enhance its solubility and intestinal permeability, leading to a significant increase in bioavailability. Both MEs showed enhanced intestinal permeability compared to free OA according to PAMPA assay results. The study suggests that MEs could be a promising formulation to improve the oral bioavailability of OA with potential clinical applications.

Summary: Usnic acid is a common metabolite of lichens with various pharmacological properties. However, its poor solubility and hepatotoxicity limit its use in therapy. In this study, polymeric micelles were developed to improve the solubility and stability of usnic acid, and it was found that the micelles exhibited inhibitory action on neuroblastoma cell migration.

Antinociceptive Action of Thymoquinone-Loaded Liposomes in an In Vivo Model of Tendinopathy

Summary: The aim of this study was to test the protective and pain reliever effect of thymoquinone (TQ)-loaded formulations in a rat model of tendinopathy induced by carrageenan intra-tendon injection. The results showed that liposomes containing 2 mg/mL of TQ and covered with hyaluronic acid (HA-LP-TQ2) reduced the development of spontaneous nociception and hypersensitivity, providing a long-lasting effect as a new treatment for tendinopathies.

Pentacyclic Triterpenes from Olive Leaves Formulated in Microemulsion: Characterization and Role in De Novo Lipogenesis in HepG2 Cells

Summary: Olive leaves contain various pentacyclic triterpenes with pharmacological activities, including antihyperlipidemic effects. A microemulsion loading a TTP-enriched extract was developed to improve the water solubility and permeability of the triterpenes. The microemulsion formulation showed high stability, prolonged release of triterpenes, and enhanced inhibitory activity on intracellular lipid accumulation.

Summary: The extensive use of agricultural pesticides increases the risk of public exposure to pesticides over time, but little attention has been paid to prevention. This study explores the potential protective effect of OleA, a component of extra virgin olive oil, against pesticide-induced cellular toxicity. The findings suggest that OleA can maintain cellular function and prevent epigenetic modification in the presence of certain pesticides.
