Effect of prolonged water stress on essential oil content, compositions and gene expression patterns of mono- and sesquiterpene synthesis in two oregano (Origanum vulgare L.) subspecies

Origanum vulgare L, recognized throughout the world as a popular medicinal and flavoring herb, contains a wide array of medicinally active components, including phenolic glucosides, flavonoids, tannins, sterols and high amounts of terpenoids. Especially the latter are often extracted by hydrodistillation resulting in the so-called essential oil that is rich in monoterpenes (e.g. carvacrol, thymol, linalyl acetate) and/or sesquiterpenes (e.g. (E)-beta-caryophyllene, germacrene D, bicyclogermacrene, 13-caryophyllene oxide). Water stresses in the arid and semiarid regions of the world severely affect growth and productivity of oregano. To determine the variation in essential oil and gene expression pathway of Iranian oregano under prolonged water stress, two native subspecies of 0. vulgare (subsp. virens and subsp. gracile) were studied. The plants, grown in pots, were subjected to three water stress conditions, i.e. no stress, mild stress (60 +/- 5% FMC) and moderate stress (40 +/- 5% FMC). The studied subspecies exhibited significant differences in essential oil content, compositions, and patterns of gene expression under water stress conditions. The essential oil of O. vulgare subsp. gracile was rich in the phenolic monoterpene carvacrol (46.86-52.07%), whereas the sesquiterpene hydrocarbon (Z)-alpha-bisabolene (39.17-42.64%) was the major constituent in the oil of O. vulgare subsp. virens. Both the mild and moderate water stresses significantly increased the essential oil content of O. vulgare subsp. gracile, but did not significantly change the essential oil content of O. vulgare subsp. virens nor the level of carvacrol and (Z)alpha-bisabolene in the investigated subspecies. Interestingly, the amount of (E(beta-caryophyllene in O. vulgare subsp. virens was significantly increased under water stress conditions. Gene expression studies supported the above findings and demonstrated that there are two different pathways affecting the biosynthesis of the terpenoid precursors geranyl pyrophosphate (GPP) and farnesyl pyrophosphate (FPP). In O. vulgare subsp. gracile, HMGR, Ovtps2 and CYP71D180 transcript were up -regulated under mild and moderate water stress conditions. Transcription of FPPS was apparently down -regulated in water stressed O. vulgare subsp. gracile. Investigation of terpene synthases expression levels in oregano subspecies demonstrated that Ovtps2 and Ovtps6 controlled the concentration of carvacrol and (E)-beta-caryophyllene in oregano essential oils, respectively. Introducing Three New Fruit-Scented Mints to Farmlands: Insights on Drug Yield, Essential-Oil Quality, and Antioxidant Properties

Summary: This study compared the yield, antioxidant properties, and essential-oil (EO) quality of different mint species. The results showed that grapefruit mint had the highest yield and EO content, while ginger mint had the highest antioxidant activity. Pineapple and grapefruit mint had the highest flavonoids content and DPPH scavenging activity. Gas chromatography analysis identified specific EO types in each mint. These findings have important implications for the cultivation of mint varieties with desired characteristics for the food and drug industries.

It was found that cumin, sea fennel, and anise essential oils were effective against various microorganisms. The antimicrobial activity was further enhanced when the essential oils were formulated into microemulsions, resulting in lower minimum inhibitory concentration and minimum bactericidal concentration values.

Stachys lavandulifolia Populations: Volatile Oil Profile and Morphological Diversity

The populations have potential for breeding programs and future conservation and exploitation initiatives.

Therefore, this integrated approach is recommended for enhancing sage production and improving its quality in drought conditions.

CmCML11 interacts with CmCAMTA5 to enhance γ-aminobutyric acid (GABA) accumulation by regulating GABA shunt in fresh-cut cantaloupe

Summary: This study investigated the effect of calcium chloride (CaCl2) treatment on GABA accumulation in fresh-cut cantaloupe and the underlying mechanisms. The results showed that CaCl2 treatment increased GABA content and the activities of GAD and SSADH enzymes, while reducing glutamate content and GABA-T activity. Additionally, CaCl2 treatment upregulated the expressions of CmCML11 and CmCAMTA5, as well as several GABA shunt genes, through the transcriptional activation by CmCAMTA5. Furthermore, the interaction between CmCML11 and CmCAMTA5 enhanced the transcriptional activation of GABA shunt genes. Overall, this study reveals that CaCl2 treatment promotes GABA accumulation in fresh-cut cantaloupe through the combined effect of CmCML11 and CmCAMTA5 in regulating the expressions of GABA shunt genes.

Integrated physiological, biochemical, and transcriptomics analyses reveal the underlying mechanisms of high nitrogen use efficiency of black sesame

Summary: This study identified the high nitrogen use efficiency (NUE) black sesame variety 17-156 and analyzed its underlying physiological and molecular mechanisms. The results showed that 17-156 possesses a sophisticated nitrogen metabolizing machinery to uptake and assimilate higher quantities of inorganic nitrogen, simultaneously improving carbon metabolism and growth. Many important genes were up-regulated in 17-156 under high nitrogen condition. Additionally, 38 potential candidate genes were identified for future studies to improve sesame's NUE. These findings provide valuable resources for understanding the regulatory network of nitrogen metabolism and developing sesame cultivars with improved NUE.

Summary: Selenium (Se) can reduce uptake and translocation of cadmium (Cd) in plants by regulating root morphology. This study investigated the effects of Se(IV) on root exudates, root morphology, root endogenous hormones, and Cd uptake efficiency in rice under Cd stress. The results showed that Se(IV) significantly reduced Cd concentrations in shoots and roots, and decreased Cd uptake efficiency via root hairs. Se(IV) also affected root morphology, root exudates, and the synthesis of hormones like IAA and JA. However, transcriptome analysis revealed no upregulated differentially expressed genes (DEGs) in IAA synthesis.
