The diagnostic role of plasma circulating precursors of miRNA-944 and miRNA-3662 for non-small cell lung cancer detection

Introduction: microRNA (miRNA) seem to be most attractive cancer markers due their crucial role in tumor development and possibility of their analysis using liquid biopsy. To date there is little known about role of miRNA precursors (pri-miRNA) in carcinogenesis and their utility as tumor markers. Material and methods: miRNA-944 and miRNA-3662 precursors as potential non-small cell lung cancer (NSCLC) markers were analyzed in plasma samples of 56 patients in an early stage of NSCLC and 100 healthy individuals. Results: Diagnostic test based on two studied markers for stage I-111A of the disease allowed to distinguish NSCLC from healthy individuals with 75.7% sensitivity and 82.3% specificity (AUC = 0.898). pri-miRNA-944 distinguished SCC from AC with sensitivity of 78.6% and specificity of 91.7% (AUC = 0.771), and pri-miRNA-3662 distinguished AC from SCC with 57.1% sensitivity and 90% specificity (AUC = 0.845). Conclusion: Circulating pri-miRNA-944 and 3662 can improve non-invasive NSCLC detection of operable stages of SCC and AC. miRNA precursors could be considered as novel potential lung cancer biomarkers.

Union is strength: Textbook outcome with perioperative chemotherapy compliance decreases the risk of death in advanced gastric cancer patients

Summary: The textbook oncological outcome (TOO) of multimodal treatment has a significant impact on the prognosis of patients with advanced gastric cancer. Patients who received multimodal treatment had lower risk of death compared to those who only received preoperative chemotherapy. Additionally, patients with pathological response in the primary tumor also had a lower risk of death. Thus, TOO can serve as an evaluation parameter for the quality of multimodal treatment in advanced gastric cancer patients.

TNF-α Induced Myotube Atrophy in C2C12 Cell Line Uncovers Putative Inflammatory-Related lncRNAs Mediating Muscle Wasting

Summary: This study aimed to evaluate the impact of inflammatory conditions on the expression of long non-coding RNAs (lncRNAs) in a mouse skeletal muscle cell line. Three lncRNAs (Gm4117, Ccdc41os1, and 5830418P13Rik) were found to be significant in inflammatory-related myotube atrophy. The expression of lncRNAs varied depending on the concentration of the inflammatory factor and the treatment time. Bioinformatics analysis identified potential target genes and miRNAs for these lncRNAs, further implicating their involvement in molecular processes related to muscle atrophy.

Summary: The aim of this study was to evaluate the levels of MMPs and osteocalcin in the serum and synovial fluid of obese women with osteoarthritis and their correlations with clinical status. The results showed that the levels of MMP3, MMP9, and pro-MMP13 were increased in more advanced stages of osteoarthritis, indicating an underlying inflammatory process.

Prognostic value of molecular cytology by one-step nucleic acid amplification (OSNA) assay of peritoneal washings in advanced gastric cancer patients

Summary: This study evaluated the prognostic value of the One-Step Nucleic Acid Amplification (OSNA) assay in advanced gastric cancer patients. The results showed that a positive OSNA assay result was a significant unfavorable prognostic factor in these patients. The OSNA assay of peritoneal washings could be considered as an adjunct tool to provide precise intraoperative staging and additional prognostic information.

Serum Calprotectin - a NET Product - as a Biomarker of Disease Activity in Patients with Systemic Lupus Erythematosus: A Single-Center Case-Control Study from Poland

Summary: This study aimed to evaluate the potential of serum calprotectin as a rapid diagnostic biomarker for systemic lupus erythematosus (SLE). Comparing 59 SLE patients with 52 healthy controls, it was found that the study group had significantly higher levels of calprotectin. Calprotectin showed high sensitivity and specificity in differentiating SLE patients from healthy volunteers, as well as in detecting moderate to severe SLE patients.

The TT genotype is linked to shorter progression-free survival (PFS) and overall survival (OS). Therefore, evaluating the SNP 686C > T of the KIAA1524 gene could serve as a diagnostic tool for identifying MM patients at risk of early disease progression and death.

The Clinical Relevance of Selected Cytokines in Newly Diagnosed Multiple Myeloma Patients

Summary: This study assessed the correlation between pretreatment serum concentrations of IL-6, IL-8, MCP-1, and VEGF and the clinical outcomes of MM patients. Results showed that IL-8 was significantly correlated with age, staging, performance status, anemia, kidney disease, and VEGF. Chemotherapy responders had lower IL-8, IL-6, and VEGF concentrations compared to non-responders. High levels of IL-6 were associated with a higher risk of disease progression and reduced overall survival.

PPY-cell hyperplasia accompanying NENs: Immunohistochemical and nuclear medicine analysis

Summary: Pancreatic polypeptide cell hyperplasia (PPY-H) is a multiplication of neuroendocrine cells producing pancreatic polypeptide (PPY). The development and role of PPY-H still need to be further elucidated. This study analyzed 12 cases of PPY-H accompanying pancreatic neuroendocrine neoplasias (NEN) and found that gastrointestinal symptoms may be more related to PPY-H rather than NEN hormonal production. Strong SSTR2 expression in PPY-H suggests the potential utility of radiotracers in clinical diagnostics, but further studies are needed.
