This study compared the antioxidant status and major lipophilic antioxidants in patients with ataxia-telangiectasia (AT) and Nijmegen breakage syndrome (NBS). Total antioxidant status (TAS), total oxidant status (TOS), oxidative stress index (OSI), and concentrations of coenzyme Q10 (CoQ10) and vitamins A and E were estimated in the plasma of 22 patients with AT, 12 children with NBS, and the healthy controls. In AT patients, TAS (median 261.7 mu mol/L) was statistically lower but TOS (496.8 mu mol/L) was significantly elevated in comparison with the healthy group (312.7 mu mol/L and 311.2 mu mol/L, resp.). Tocopherol (0.8 mu g/mL) and CoQ10 (0.1 mu g/mL) were reduced in AT patients versus control (1.4 mu g/mL and 0.3 mu g/mL, resp.). NBS patients also displayed statistically lower TAS levels (290.3 mu mol/L), while TOS (404.8 mu mol/L) was comparable to the controls. We found that in NBS patients retinol concentration (0.1 mu g/mL) was highly elevated and CoQ10 (0.1 mu g/mL) was significantly lower in comparison with those in the healthy group. Our study confirms disturbances in redox homeostasis in AT and NBS patients and indicates a need for diagnosing oxidative stress in those cases as a potential disease biomarker. Decreased CoQ10 concentration found in NBS and AT indicates a need for possible supplementation.

Insights into Prevention of Health Complications in Small for Gestational Age (SGA) Births in Relation to Maternal Characteristics: A Narrative Review

Sebastian Wolejszo, Agnieszka Genowska, Radoslaw Motkowski, Birute Strukcinskiene, Mark Klukowski, Jerzy Konstantynowicz

Summary: This review aims to summarize maternal biological and socio-demographic factors as well as preventive strategies to reduce the risk of Small for Gestational Age (SGA) births. Current evidence shows that factors such as primiparity, previous stillbirths, maternal age, single motherhood, low socio-economic status, smoking, and cannabis use during pregnancy significantly increase the risk of SGA births. Beneficial factors include the Mediterranean diet and dietary intake of vegetables, while studies on alcohol consumption during pregnancy and SGA birth weight are inconclusive. A midwifery-led model based on early and extensive prenatal care has been shown to reduce the risk of SGA births. It is important to educate parents about modifiable and non-modifiable risk factors and implement preventive strategies for SGA births.

Lipid peroxidation and sphingolipid alterations in the cerebral cortex and hypothalamus of rats fed a high-protein diet

Elzbieta Supruniuk, Ewa Zebrowska, Mateusz Maciejczyk, Anna Zalewska, Adrian Chabowski

Summary: This study examined the effects of high-protein diets on lipid peroxidation and sphingolipid metabolism in rats. The results showed that high-protein diets significantly impacted hypothalamic metabolic pathways, leading to increased lipid peroxidation, ceramide accumulation, and changes in sphingolipid metabolism.

Summary: Chronic kidney disease (CKD) is a systemic inflammatory disease that affects multiple organs, including the kidneys, cardiovascular system, and oral cavity. Children with CKD experience reduced saliva secretion (hyposalivation), leading to increased dental caries and impaired quality of life. This study evaluated the inflammatory and anti-inflammatory profile in the saliva of CKD children at different stages of renal failure. The results showed significant changes in salivary cytokines, chemokines, and growth factors in CKD children compared to healthy controls.

Diagnostic significance and utility of circulating redox biomarkers in patients with gastric cancer - preliminary study

Justyna Dorf, Konrad Zareba, Anna Pryczynicz, Joanna Matowicka-Karna, Boguslaw Kedra, Piotr Zukowski, Anna Zalewska, Mateusz Maciejczyk

Summary: Gastric cancer is associated with redox imbalance and increased oxidative damage to proteins, lipids, and DNA. Histopathological parameters of the tumor are associated with the level of antioxidants and oxidative damage products. Determination of redox parameters may be useful in the assessment of the tumor progression.

Summary: Although titanium and its alloys have high biocompatibility, there is a growing debate regarding the removal of titanium implants due to potential long-term adverse effects. To enhance the biocompatibility of titanium implants, increasing the thickness of the titanium dioxide passive layer is being explored. In this study, we compared the cytotoxicity, mitochondrial function, and redox balance of fibroblast mitochondria between hard-anodized (type II), standard-anodized (type III), and non-anodized titanium discs. The results showed oxidative stress in fibroblast mitochondria regardless of anodization, but type II anodization prevented changes in complex II activity, had the lowest citrate synthase inhibition, and reduced cytochrome c oxidase inhibition, potentially preventing apoptosis.

IMMUNOAGING - THE EFFECT OF AGE ON SERUM LEVELS OF NET BIOMARKERS IN MEN: A PILOT STUDY

Marzena Garley, Wioleta Justyna Omeljaniuk, Radoslaw Motkowski, Wioletta Ratajczak-Wrona, Ewa Jablonska, Daniel Filipkowski, Angelika Edyta Charkiewicz

Summary: This study evaluated the impact of aging on the formation of neutrophil extracellular traps (NETs). The results showed that levels of myeloperoxidase (MPO) and circulating free DNA (cfDNA), which are biomarkers of NETs, increased with age until adulthood, but were lower in older men, suggesting impaired NET formation.

Dariusz Juchnowicz, Michal Dzikowski, Joanna Rog, Napoleon Waszkiewicz, Kaja Hanna Karakula, Anna Zalewska, Mateusz Maciejczyk, Hanna Karakula-Juchnowicz

Summary: This study assessed the utility of complete blood count (CBC) in the prediction of schizophrenia diagnosis and its relationship with oxidative stress (OS). The findings showed that certain CBC parameters were altered in patients with the first episode of psychosis compared to healthy individuals, and there were various associations between OS and CBC. The study also proposed promising CBC models for distinguishing different patient groups.

Summary: This study analyzed the concentrations of renal biomarkers (osteopontin, cystatin C, and NGAL) in neonates and found that prematurity may lead to increased excretion of osteopontin and cystatin C, but not NGAL. The excretion of NGAL and cystatin C may change during the first weeks of a premature neonate's life, but osteopontin does not.

Summary: This study compares the antioxidant and antiglycation potentials of popular H2 antagonists (ranitidine, cimetidine, and famotidine) on protein glycoxidation. The results show that ranitidine is the only H2 blocker that significantly inhibits protein glycation and has antioxidant properties. It can compete with proteins for the addition of carbonyl groups.

Preliminary Assessment of Polysaccharide-Based Emulgels Containing Delta-Aminolevulinic Acid for Oral Lichen planus Treatment

Emilia Szymanska, Joanna Potas, Mateusz Maciejczyk, Magdalena Ewa Sulewska, Malgorzata Pietruska, Anna Zalewska, Aleksandra Pietruska, Katarzyna Winnicka

Summary: This study tested different emulgel compositions for the delivery of delta-aminolevulinic acid and investigated the impact of the drug on the behavior of gelling agents. The results showed that a combination of xanthan and gellan gum enhanced the retention of the emulgels in oral mucosal tissue. In addition, the incorporation of delta-aminolevulinic acid promoted interactions with mucosal tissue. Cytotoxicity studies demonstrated that emulgels based on tragacanth and gellan/xanthan may have a protective effect on the metabolic activity of human gingival fibroblasts and keratinocytes.
