Insulin-Like Growth Factor (IGF) Binding Protein-2, Independently of IGF-1, Induces GLUT-4 Translocation and Glucose Uptake in 3T3-L1 Adipocytes

Insulin-like growth factor binding protein-2 (IGFBP-2) is the predominant IGF binding protein produced during adipogenesis and is known to increase the insulin-stimulated glucose uptake (GU) in myotubes. We investigated the IGFBP-2-induced changes in basal and insulin-stimulated GU in adipocytes and the underlying mechanisms. We further determined the role of insulin and IGF-1 receptors in mediating the IGFBP-2 and the impact of IGFBP-2 on the IGF-1-induced GU. Fully differentiated 3T3-L1 adipocytes were treated with IGFBP-2 in the presence and absence of insulin and IGF-1. Insulin, IGF-1, and IGFBP-2 induced a dose-dependent increase in GU. IGFBP-2 increased the insulin-induced GU after long-term incubation. The IGFBP-2-induced impact on GU was neither affected by insulin or IGF-1 receptor blockage nor by insulin receptor knockdown. IGFBP-2 significantly increased the phosphorylation of PI3K, Akt, AMPK, TBC1D1, and PKC zeta/lambda and induced GLUT-4 translocation. Moreover, inhibition of PI3K and AMPK significantly reduced IGFBP-2-stimulated GU. In conclusion, IGFBP-2 stimulates GU in 3T3-L1 adipocytes through activation of PI3K/Akt, AMPK/TBC1D1, and PI3K/PKC zeta/lambda/GLUT-4 signaling. The stimulatory effect of IGFBP-2 on GU is independent of its binding to IGF-1 and is possibly not mediated through the insulin or IGF-1 receptor. This study highlights the potential role of IGFBP-2 in glucose metabolism.

Summary: The objective of this study was to examine the effects of lifestyle intervention on impaired insulin secretion in specific subphenotypes of prediabetes. Participants from the Prediabetes Lifestyle Intervention Study were divided into six prediabetes clusters and assessed for insulin secretion, glycemia, insulin sensitivity, and liver fat changes during a 1-year intervention. The lifestyle intervention improved glycemic traits in all high-risk clusters, while insulin secretion increased only in clusters 3 and 5. However, high liver fat in cluster 5 hindered the improvement of insulin secretion.

The role of hepatokines in NAFLD

Summary: Non-alcoholic fatty liver disease (NAFLD) is both a consequence and a cause of insulin resistance and major non-communicable diseases (NCDs). The close relationship between NAFLD and visceral obesity has led to the overshadowing of fatty liver as the main pathomechanism of insulin resistance and NCDs. However, the concept of hepatokines provides a new perspective on the role of the liver in metabolism and offers potential for precision medicine in NAFLD and visceral obesity.

Implication of sugar, protein and incretins in excessive glucagon secretion in type 2 diabetes after mixed meals

Circulating levels of gastrointestinal hormones in prediabetes reversing to normoglycemia or progressing to diabetes in a year-A cross-sectional and prospective analysis

Summary: This study aimed to compare the concentrations of GLP-1, glucagon, GIP, and glicentin during an OGTT in patients with different glycemic states. The results showed that hormonal levels did not differ between groups when all subjects were in a prediabetic state. However, one year later, patients progressing to diabetes had lower postprandial increases of glicentin and GLP-1, lower postprandial decrease of glucagon, and higher levels of fasting GIP compared to patients regressing to NGT. Changes in glicentin and GLP-1 were found to be negatively correlated with changes in glucose levels and beta-cell function markers.

Ex vivo instability of lipids in whole blood: preanalytical recommendations for clinical lipidomics studies

Summary: Reliability, robustness, and interlaboratory comparability of quantitative measurements in clinical lipidomics studies are crucial. The different ex vivo stability of lipids in blood poses a risk to data interpretation. Clear recommendations for blood sample collection are necessary. In this study, we used UHPLC-high resolution mass spectrometry to investigate the stability of 417 lipid species in EDTA whole blood under different temperature conditions and time points. Our data suggest that cooling whole blood immediately and continuously, and separating plasma within 4 hours, unless focusing on robust lipids, are recommended. Lists of lipids' ex vivo (in)stability and potential biomarkers in whole blood are provided. Overall, our results contribute to the international efforts in achieving reliable and comparable clinical lipidomics studies in the future.

Summary: This study demonstrates that the ATP citrate lyase inhibitor Bempedoic acid can significantly reduce low-density lipoprotein cholesterol levels and is associated with a low incidence of muscle-related adverse events. Among patients unable or unwilling to take statins, Bempedoic acid treatment significantly reduces the risk of major adverse cardiovascular events.
