Many reports have revealed the importance of the sympathetic nervous system (SNS) in the control of the bone marrow environment. However, the specific role of neuropeptide Y (NPY) in this process has not been systematically studied. Here we show that NPY-deficient mice have significantly reduced hematopoietic stem cell (HSC) numbers and impaired regeneration in bone marrow due to apoptotic destruction of SNS fibers and/or endothelial cells. Furthermore, pharmacological elevation of NPY prevented bone marrow impairments in a mouse model of chemotherapy-induced SNS injury, while NPY injection into conditional knockout mice lacking the Y1 receptor in macrophages did not relieve bone marrow dysfunction. These results indicate that NPY promotes neuroprotection and restores bone marrow dysfunction from chemotherapy-induced SNS injury through the Y1 receptor in macrophages. They also reveal a new role of NPY as a regulator of the bone marrow microenvironment and highlight the potential therapeutic value of this neuropeptide.

nNOS regulates ciliated cell polarity, ciliary beat frequency, and directional flow in mouse trachea

Anatoly Mikhailik, Tatyana Michurina, Krikor Dikranian, Stephen Hearn, Vladimir Maxakov, Saul S. Siller, Ken-Ichi Takemaru, Grigori Enikolopov, Natalia Peunova

Summary: This passage discusses the crucial role of nitric oxide in regulating the polarity and ciliary beat frequency of multiciliated cells in the airway, and how this impacts airway flow. It also suggests a potential explanation for the low levels of exhaled nitric oxide in ciliopathies due to deficiencies in NO availability in ciliated cells.

Lack of peptide YY signaling in mice disturbs gut microbiome composition in response to high-fat diet

Aitak Farzi, Chi Kin Ip, Felicia Reed, Ronaldo Enriquez, Geraldine Zenz, Marija Durdevic, Lei Zhang, Peter Holzer, Herbert Herzog

Summary: The lack of PYY in mice results in distinct changes in gut microbiome composition, especially under high-fat diet conditions. Pyy-KO mice showed an increased abundance of Bacteroidetes phylum and specific genera within it, indicating a gene-environment interaction between diet and gut microbiome that can impact gut homeostasis.

Zap70 Regulates TCR-Mediated Zip6 Activation at the Immunological Synapse

Bonah Kim, Hee Young Kim, Won-Woo Lee

Summary: Zinc, an essential microelement, plays immunoregulatory roles by influencing signaling pathways to enhance immune function. Zinc deficiency impairs immunity and increases susceptibility to infection. The interaction between Zip6-mediated zinc influx and TCR activation regulates T-cell responses.

The bioavailability time of commonly used thymidine analogues after intraperitoneal delivery in mice: labeling kinetics in vivo and clearance from blood serum

Summary: The detection of synthetic thymidine analogues incorporated into replicating DNA during the S-phase of the cell cycle is a valuable methodology for evaluating proliferative activity. Recent studies have shown that different thymidine analogues exhibit similar labeling kinetics and clearance rates from blood serum, with a bioavailability time of approximately 1 hour in vivo. This finding is significant for the practical use of multiple S-phase labeling with various analogues.

Summary: This study introduces an algorithm called CORGI for the registration of perinatal brains. The algorithm utilizes image preprocessing, attention-gated simulated annealing, and classical multidimensional scaling to improve the accuracy of registration. Tested on samples of perinatal mouse brains, the algorithm shows promising results and automates brain registration tasks with a short runtime, making it significant for studying brain development dynamics.

Sustained glymphatic transport and impaired drainage to the nasal cavity observed in multiciliated cell ciliopathies with hydrocephalus

Summary: This study investigated the impact of ciliopathies on CNS fluid homeostasis. The results showed that sustained glymphatic transport and impaired solute drainage via the cribriform plate to the nasal cavity were observed in models of ciliopathy, along with the occurrence of hydrocephalus. This research is important for understanding how ciliopathies contribute to disruption of CNS fluid homeostasis.

Sex-specific changes in metabolism during the transition from chow to high-fat diet feeding are abolished in response to dieting in C57BL/6J mice

Jennifer Oraha, Ronaldo F. Enriquez, Herbert Herzog, Nicola J. Lee

Summary: This study found that C57BL/6J mice exhibit sex-specific behavioral and metabolic responses to the introduction and sustained intake of a high-fat diet (HFD), but consistent responses to a dieting situation. Males showed immediate and significant weight gain, while females displayed stronger resistance to the obesifying effects of HFD. This sex disparity may be attributed to inherent sex differences in neurological signaling pathways and fat utilization. Interestingly, both sexes showed similar responses to returning to a normal diet.

Summary: This study examines the effects of thymidine analogs on adult hippocampal neurogenesis and perinatal somatic development of mice. Detection of De Novo Dividing Stem Cells In Situ through Double Nucleotide Analogue Labeling

Sheed Itaman, Grigori Enikolopov, Oleg V. Podgorny

Summary: Tissue-specific somatic stem cells are characterized by their ability to be in a state of prolonged reversible cell cycle arrest. Maintaining a balance between cell quiescence and division is crucial for tissue homeostasis. However, detecting de novo dividing cells in quiescent stem cells has been challenging. In this study, a novel method based on double labeling with thymidine analogues was presented and experimentally verified to detect de novo dividing stem cells in situ.

Anatomical Consideration for Anterior Approach of Cervicothoracic Junction: A Computed Tomography Image Analysis

Eugene J. Park, Bo-Gil Jeong, Woo-Kie Min

Summary: This study analyzed the anatomical structures encountered during anterior approaches in the cervicothoracic junction (CTJ) and evaluated the feasibility of previously reported surgical corridors. The intercarotid artery angle (ICAA) and intercarotid artery distance (ICAD) were larger in men. The shape of the brachiocephalic trunk (BCT) was often convex and located inside the body. The accessible levels for anterior approaches were T1, T3, and T5. Preoperative analysis of vascular structures and accessible levels is essential for deciding the appropriate surgical corridor and reducing complications.
