In early 2016, the FDA and the National Institutes of Health (NIH) published the first version of the glossary included in the Biomarkers, EndpointS, and other Tools (BEST) resource. 1 The BEST glossary was constructed to harmonize and clarify terms used in translational science and medical product development and to provide a common language used for communication by those agencies. It is considered a living document that will be updated in the future. This review will discuss the main biomarker and clinical outcome categories contained in the BEST glossary as they apply to neuro-oncology, as well as the overlapping and hierarchical relationships among them.

Summary: The interferon gene signature (IGS) in early, treatment naive rheumatoid arthritis (eRA) patients can negatively affect initial treatment response by causing sustained, epigenetically mediated increases in lymphocyte activation and proliferation. This highlights the importance of the IGS as a prognostic biomarker and suggests the rationale for targeting IFN-alpha in selected eRA patients.

Summary: This study aimed to sensitize colorectal cancer cells to chemotherapy by targeting the IRE1a/XBP1 pathway. The results showed that the IRE1a inhibitor 4 mu 8C inhibited CRC cell growth and enhanced the cytotoxicity of 5-FU. In vivo experiments demonstrated that 4 mu 8C inhibited tumor growth and significantly enhanced drug responses when combined with 5-FU.

Facial pain and sensory outcomes following resection of tumors compressing the trigeminal nerve

Summary: Most patients with facial sensory symptoms caused by meningiomas or vestibular schwannomas (VSs) experienced improvement after tumor resection. Surgery led to immediate and sustained improvement in pain and paresthesias, whereas numbness was slower to improve. Patients with smaller tumors and complete decompression of the trigeminal nerve were more likely to experience improvement in facial pain.
