Activation of Nrf2 Signaling Augments Vesicular Stomatitis Virus Oncolysis via Autophagy-Driven Suppression of Antiviral Immunity

Oncolytic viruses (OVs) offer a promising therapeutic approach to treat multiple types of cancer. In this study, we show that the manipulation of the antioxidant network via transcription factor Nrf2 augments vesicular stomatitis virus Delta 51 (VSV Delta 51) replication and sensitizes cancer cells to viral oncolysis. Activation of Nrf2 signaling by the antioxidant compound sulforaphane (SFN) leads to enhanced VSV Delta 51 spread in OV-resistant cancer cells and improves the therapeutic outcome in different murine syngeneic and xenograft tumor models. Chemoresistant A549 lung cancer cells that display constitutive dominant hyperactivation of Nrf2 signaling are particularly vulnerable to VSV Delta 51 oncolysis. Mechanistically, enhanced Nrf2 signaling stimulated viral replication in cancer cells and disrupted the type I IFN response via increased autophagy. This study reveals a previously unappreciated role for Nrf2 in the regulation of autophagy and the innate antiviral response that complements the therapeutic potential of VSV-directed oncolysis against multiple types of OV-resistant or chemoresistant cancer.

Summary: The long non-coding RNA LUCAT1 is a negative feedback regulator of interferon and cytokine expression in human myeloid cells. It binds to proteins involved in mRNA processing and alternative splicing, and its absence leads to altered splicing and reduced expression of the NR4A2 gene, resulting in elevated immune gene expression. Furthermore, elevated LUCAT1 levels are found in patients with inflammatory bowel disease, asthma, and chronic obstructive pulmonary disease, suggesting its role in the regulation of inflammatory diseases.

Summary: Hyperexcitability is associated with neuronal dysfunction, cellular death, and neurodegeneration. NOX4, an NADPH oxidase, is found to regulate reactive oxygen species (ROS) levels and calcium homeostasis, thereby preventing hyperexcitability and neuronal death. These findings suggest that NOX4 might have a crucial role in neurodegenerative diseases by acting as a potential redox regulator.

Nrf2 regulates glucose uptake and metabolism in neurons and astrocytes

Summary: The transcription factor Nrf2 and its repressor Keap1 play a crucial role in cell stress adaptation by regulating gene expression related to cellular detoxification, antioxidant defense, and energy metabolism. This study investigated the impact of Nrf2 on glucose distribution and the connection between NADH production in energy metabolism and NADPH homeostasis using glio-neuronal cultures. The findings showed that Nrf2 activation enhances glucose uptake in neurons and astrocytes, with prioritized consumption for mitochondria-related energy production rather than NADPH synthesis in the pentose phosphate pathway.

Summary: The study evaluated the effect of swietenine in nonalcoholic fatty liver disease (NAFLD) in diabetic mice. Swietenine was found to reverse hepatic lipid accumulation and activate the Nrf2 antioxidant defense pathway, inhibiting lipogenesis. These findings suggest that swietenine has beneficial effects in diabetes-induced NAFLD.

Nrf2 in TIME: The Emerging Role of Nuclear Factor Erythroid 2-Related Factor 2 in the Tumor Immune Microenvironment

Summary: Nuclear factor erythroid 2-related factor 2 (Nrf2) mediates cellular antioxidant response and plays a role in metabolism, inflammation, and immunity. Nrf2 activation is often exploited by cancer cells for survival and resistance to therapies. It can also occur in tumor-associated macrophages (TAMs) and contribute to an immunosuppressive tumor immune microenvironment (TIME). Cancer cell-derived metabolites activate Nrf2 and modulate the interaction between TIME and TAMs. Understanding the effects of Nrf2 in cancer presents opportunities for targeted therapy.
