Summary: This study compared the efficacy of different induction regimens in patients with initially unresectable colorectal cancer liver metastases. The results showed that FOLFOXIRI-bevacizumab was the preferred treatment for patients with right-sided primary tumor or RAS/BRAFV600E mutation, while the addition of panitumumab to FOLFOX or FOLFIRI in patients with left-sided and RAS/BRAFV600E wild-type tumor showed no clinical benefit but more toxicity.

Summary: RAS mutations are prognostic for mCRC patients. KRAS exon 3/4 mutations are rare and may be associated with a more favorable prognosis, but the difference in oncologic outcomes based on the mutated exon appears to be smaller than previously reported.

Summary: Strong immune responses in primary colorectal cancer are associated with better patient survival. However, identifying patients with good prognosis after surgery for colorectal cancer liver metastases remains challenging. This study aims to determine the practical application of immune cell infiltration and stromal content in predicting outcome and investigate the underlying functional biology.

Summary: This study compared the histopathological responses of liver-limited colorectal metastases in patients who received different chemotherapy regimens and found differences between bevacizumab and cetuximab in terms of histological response. The study also showed that tumor regression grade (TRG) and dangerous halo (DH) are associated with prognosis.

Summary: The study demonstrates that the addition of TARE to second-line chemotherapy for CLM patients results in longer progression-free survival and hepatic progression-free survival, with an improved objective response rate. However, there was no significant difference in overall survival between the two groups, and the rate of grade 3 adverse events was higher in the TARE group. Further subset analyses are needed to identify the ideal patient population for TARE benefit.

Summary: The liver is a common site for metastasis in colorectal cancer patients due to the direct connection between the colon and rectum and the liver through the portal vein circulation. The liver tumor microenvironment consists of different cell types that modulate antigen recognition and immune system activation. Primary tumors from other sites create a pre-metastatic niche in the liver before the seeding of cancer cells. There are four key phases in the development of liver metastases: microvascular infiltration, extravascular pre-angiogenesis, angiogenesis, and tumor growth.

Summary: A predictive model for conversion surgery was developed and the long-term outcomes of patients with technically unresectable colorectal liver metastases were evaluated. Patients who underwent conversion surgery had significantly higher median overall survival compared to those who did not. The predictive model for conversion surgery was constructed using several predictive factors. Patients were graded according to their scores, and those in Grade A had significantly better overall survival than Grade B and C patients.

Summary: Adding von Willebrand factor antigen (vWF-Ag) and C-reactive protein (CRP) to the Model for End-Stage Liver Disease-Sodium (MELD-Na) score improves risk stratification of patients awaiting liver transplant, especially in predicting waitlist mortality. The combination of vWF-Ag and CRP provides better predictive value for waitlist mortality, reflecting central pathophysiological mechanisms of portal hypertension, bacterial translocation, and inflammation.

Summary: This study found that the use of a selective SHP1/SHP2 inhibitor, NSC-87877, can accelerate hepatocyte proliferation, improve liver regeneration, and reduce injury after hepatectomy. It also demonstrated the dependence of these effects on YAP, highlighting the role of these molecules in the enhanced regenerative response. Furthermore, NSC-87877 treatment was beneficial in a murine model of nonalcoholic steatohepatitis.

Summary: This study explored the potential of circulating apoptosis markers in predicting treatment response in patients with colorectal cancer liver metastasis. The findings indicated that CRCLM tissues had more apoptotic cells and higher levels of circulating M30, which correlated with tumor volume. Patients with higher M30 levels before therapy had worse prognosis. The predictive potential of M30 was higher than that of the tumor markers CA19-9 or CEA.
