Summary: The expression of ERFE and GDF15 in MDS patients was found to be highly aberrant, with ERFE expression unexpectedly relevant for MDS prognosis. ERFE overexpression predicted superior overall survival in certain MDS subgroups, suggesting a possible involvement of ERFE in the molecular pathogenesis of MDS.

Summary: A nanochelator system utilizing the physiological environment of iron overload was developed to improve iron elimination and pharmacokinetic properties in mice, showing promising results for iron chelation therapy with no adverse effects observed in toxicity studies. This ROS-responsive nanochelator design could be a safe alternative for effectively removing iron chelates from the body in iron chelation therapy regimens requiring repeated dosing.

Summary: HIFs are critical transcription factors in the adaptive response to hypoxia and are implicated in the dysregulated immune response to various insults. However, there is a lack of studies investigating the role of HIFs in clinically relevant injury models, especially involving the lungs. This review summarizes the effects of HIF-1 alpha on the vasculature, metabolism, inflammation, and apoptosis in the lungs and discusses its role in direct lung injuries, including lung contusion, acid aspiration, pneumonia, and COVID-19.

Summary: Iron overload leads to insulin resistance in H9c2 cardiomyoblast cells. Overexpression of MitoNEET can protect against iron accumulation in the mitochondria and subsequent insulin resistance. Iron overload increases mitochondrial iron content, ROS production, mitochondrial fission, and decreases insulin-stimulated Akt and ERK1/2 phosphorylation. MitoNEET overexpression attenuates these effects and upregulates levels of PGC1 alpha protein.

Summary: Excessive production of mitochondrial reactive oxygen species (ROS) in diabetes complications is mainly caused by impaired responses to hypoxia due to the inhibition of hypoxia-inducible factor-1 (HIF-1) by hyperglycemia. Restoring HIF-1 function can attenuate ROS overproduction, protect cells against apoptosis and renal injury in diabetes. These findings suggest that targeting HIF-1 repression could be a potential therapeutic strategy for diabetic complications.

Summary: Iron overload leads to cardiovascular diseases, including heart failure. This study showed that iron overload triggers cell death in H9c2 cells by promoting intracellular and mitochondrial iron accumulation and ROS production. The use of iron-binding fluorescent reporters and antioxidants reduced these effects, and decreased apoptosis and lytic cell death. Overexpression of the mitochondrial membrane protein MitoNEET lowered iron and ROS accumulation in the mitochondria, leading to reduced cell death in response to iron overload.

Summary: Mild hypoxia attenuates the pluripotency-promoting PI3K/AKT signaling pathway in human pluripotent stem cells compared to normoxic conditions, due to the downregulation of ROS. In addition, the MAPK pathway also downregulates ROS and attenuates the PI3K/AKT signaling via a novel interaction between these pathways. This ROS-mediated regulatory circuit may have implications for enhancing cultivation and differentiation protocols for hPSCs, as well as providing insights into cancer stem cell signaling regulation.
