SUSD3 rs76292544 was associated with 12-month bleeding events and five SNPs (NCMAP rs4553122, PRF1 rs885821, PRKAG2 rs12703159, rs13224758, and POU2F3 rs2298579) were associated with peak anti-FXa level.

Summary: The study aimed to compare the odds of bleeding between rivaroxaban, enoxaparin, and aspirin following lower extremity joint arthroplasty or revision. The results showed that aspirin had significantly decreased odds of bleeding complications compared to enoxaparin, and a trend toward decreased odds compared to rivaroxaban. This suggests that aspirin may be a safer alternative for postoperative thromboprophylaxis in lower extremity joint arthroplasty or revision.

The results showed that the DL models had comparable performance with expert radiologists' PI-RADS assessment based on mpMRI, and the PIDL-CS model combining PI-RADS and DL-CS had superior performance. This study demonstrated the potential of DL models as effective auxiliary tools for predicting prostate cancer.

Summary: This study aimed to assess the feasibility of switching from dual antiplatelet therapy (DAPT) to dual pathway inhibition (DPI) and compare the pharmacodynamic (PD) profiles of these treatments. The results showed that switching from various DAPT regimens to DPI was feasible in patients with chronic coronary syndrome (CCS), with enhanced P2Y(12) inhibition with DAPT and reduced thrombin generation with DPI. There were no differences in platelet-mediated global thrombogenicity between DPI and ticagrelor- and prasugrel-based DAPT, but differences were observed with clopidogrel-based DAPT.

Summary: This study evaluated the influence of antiplatelet and anticoagulant therapy on post-operative bleeding risk in dermatosurgery. Although these medications were associated with a significantly higher rate of bleeding, no life-threatening bleeding was recorded. Prolonged preoperative pause or bridging of direct oral anticoagulants did not significantly reduce the occurrence of severe bleeding events.

The study found a lower risk of all-cause mortality in ticagrelor users compared to clopidogrel users, but no difference in other clinical endpoints. No differences were observed between prasugrel and clopidogrel. Further research is needed to determine the optimal P2Y12 inhibitor in a real-world population.

Summary: This study aimed to evaluate the safety and efficacy of different dual antiplatelet therapy (DAPT) combinations in patients with acute coronary syndrome (ACS) based on their baseline ischemic and bleeding risk. The results showed that for patients with low-to-moderate bleeding risk, clopidogrel may be associated with a higher risk of recurrent myocardial infarction (re-AMI) and increased risk of death and re-AMI in those with high ischemic risk compared to prasugrel or ticagrelor. However, there was no difference in the risk of major bleeding. Among patients with high bleeding risk, clopidogrel showed comparable risk of death, re-AMI, and major bleeding compared to potent P2Y12 inhibitors regardless of the baseline ischemic risk.

Summary: This study compared the benefits of strict vs. standard control of blood pressure with the potential benefits of controlling other modifiable risk factors in patients with chronic coronary syndromes. The results showed that the benefit associated with strict blood pressure control was marginal and driven only by the diastolic component, while uncontrolled other risk factors were associated with a gradually increasing risk. Therefore, optimizing other risk factors should be prioritized over further reduction of systolic blood pressure. The optimal blood pressure target in chronic coronary syndromes may be <=139/79 mmHg.
