Summary: Human T lymphotropic virus-associated myelopathy/tropical spastic paraparesis (HTLV/TSP) and multiple sclerosis (MS) are chronic diseases of the central nervous system, showing similarities in clinical manifestation and immunopathogenesis. Excessive immune response to the virus and neurons are the causative agents of HAM/TSP and MS. Th17 cells, a component of the immune system, play a pivotal role in the demyelination of MS and may also have a role in HAM/TSP.

There is partial symmetry in spasticity severity between the lower limbs of the individuals.

Summary: This study reveals the negative regulatory role of Yes-associated protein (YAP) in Tax activation of HTLV-1 transcription. YAP interacts with Tax and prevents the association between Tax and p300, resulting in the inhibition of HTLV-1 transcription. This finding is important for understanding the mechanisms underlying the persistent infection and latency of HTLV-1 in host cells.

Summary: Human T-cell leukemia virus type 1 (HTLV-1) is a replication-competent human retrovirus that is associated with two types of diseases, adult T-cell leukemia-lymphoma (ATL) and HTLV-1-associated myelopathy/tropical spastic paraparesis (HAM/TSP). Although most infected individuals remain asymptomatic, a subpopulation develops these diseases. The complex virus-host interactions and the host immune response may regulate the development of HTLV-1-associated diseases.

Summary: This study demonstrates that analyzing TCR repertoires of HTLV-1-infected cells can provide insights into the clonal dynamics and potential pathogenic clones expanded in the CNS of HAM/TSP patients. The TCR beta repertoire analysis reveals a higher clonal expansion in HTLV-1-infected cells compared to noninfected cells, and the TCR sequences of infected cells remain stable over time. Furthermore, highly expanded infected clones are found in the cerebrospinal fluid of HAM/TSP patients.

EZH1/2 selective inhibitors and EZH1/2 inhibitors suppressed the proliferation of HTLV-1-infected cells in HAM by inducing apoptosis and modulating immune response, suggesting the potential of these inhibitors as therapeutic agents for HAM.
