Mesenchymal stem cells from sternum: the type of heart disease, ischemic or valvular, does not influence the cell culture establishment and growth kinetics

Background: In an attempt to increase the therapeutic potential for myocardial regeneration, there is a quest for new cell sources and types for cell therapy protocols. The pathophysiology of heart diseases may affect cellular characteristics and therapeutic results. Methods: To study the proliferative and differentiation potential of mesenchymal stem cells (MSC), isolated from bone marrow (BM) of sternum, we made a comparative analysis between samples of patients with ischemic (IHD) or non-ischemic valvular (VHD) heart diseases. We included patients with IHD (n = 42) or VHD (n = 20), with average age of 60 years and no differences in cardiovascular risk factors. BM samples were collected (16.4 +/- 6 mL) and submitted to centrifugation with Ficoll-Paque, yielding 4.5 +/- 1.5 x 10(7) cells/mL. Results: Morphology, immunophenotype and differentiation ability had proven that the cultivated sternal BM cells had MSC features. The colony forming unit-fibroblast (CFU-F) frequency was similar between groups (p = 0.510), but VHD samples showed positive correlation to plated cells vs. CFU-F number (r = 0.499, p = 0.049). The MSC culture was established in 29% of collected samples, achieved passage 9, without significant difference in expansion kinetics between groups (p > 0.05). Dyslipidemia and the use of statins was associated with culture establishment for IHD patients (p = 0.049 and p = 0.006, respectively). Conclusions: Together, these results show that the sternum bone can be used as a source for MSC isolation, and that ischemic or valvular diseases do not influence the cellular yield, culture establishment or in vitro growth kinetics.

Summary: This study evaluated the indirect effects of the COVID-19 pandemic on the care of women with pregnancies complicated by gestational or pre-existing diabetes, and their maternal-fetal outcomes. The study found that despite the impact of the pandemic on the economy, society, and structure, parameters of maternal care were similar, diabetes control improved, and neonatal hypoglycemia and SGA rates were lower among pregnant women with diabetes during the pandemic.

Summary: In this study, metagenomics was used to identify viral infections among pediatric patients with acute respiratory disease. Two pools of samples were found to contain the little-known Malawi polyomavirus (MWPyV). Further testing revealed that MWPyV-positive samples also tested positive for respiratory syncytial virus. Phylogenetic analysis of MWPyV genome sequences from index samples was conducted, providing insights into its viral origin. This study represents the first molecular and genomic characterization of MWPyV in pediatric patients in South America.

Summary: A study examined the composition of plasma virome in patients with prostate cancer, finding that Anelloviruses and Human Pegivirus-1 (HPgV-1) were the most abundant components. Clinically important viruses like hepatitis C virus (HCV), cytomegalovirus, and human adenovirus type C were also identified. In comparison, the virome of blood donors was mainly composed of torque teno virus types (TTV). This study sheds light on the viral abundance and diversity in patients with prostate cancer, suggesting that malignant processes may influence virome composition.

Summary: This study aimed to investigate the associations between movement behaviors, abdominal obesity, and inflammatory biomarkers in adolescents, as well as the mediating role of waist circumference. The results showed a negative association between sleep duration and inflammatory biomarkers, and waist circumference partially mediated this association. Therefore, maintaining healthy sleep habits can have positive implications for reducing waist circumference and inflammatory indicators in adolescents.

Molecular identification of the emerging Human Gemykibivirus-2 (HuGKV-2) among Brazilian blood donors

Summary: This study investigated the prevalence of HuGkV-2 DNA among healthy blood donors from different regions in Brazil. Although this provides preliminary insights into the impact of this emerging viral agent, further studies are needed to understand the prevalence, transmission routes, and possible clinical effects.

Cardiorespiratory fitness in individuals with type 2 diabetes mellitus: a systematic review and meta-analysis
