Sustained release of parathyroid hormone via in situ cross-linking gelatin hydrogels improves the therapeutic potential of tonsil-derived mesenchymal stem cells for hypoparathyroidism

Biomimetic parathyroid regeneration with sustained release of parathyroid hormone (PTH) into the blood stream is a considerable challenge in hypoparathyroidism treatment. We recently reported that tonsil-derived mesenchymal stem cells (TMSCs), if these cells were both differentiated in vitro before implantation and incorporated into a scaffold Matrigel, are a good cell source for parathyroid regeneration in a parathyroidectomized (PTX) animal model. Here, we present a new strategy for improved clinical application that enhances the sustained release of PTH by controlling mechanical stiffness using in situ-forming gelatin-hydroxyphenyl propionic acid (GH) hydrogels (GHH). Differentiated TMSCs (dTMSCs) embedded in a GHH with a strength of 4.4kPa exhibited the best sustained release of PTH and were the most effective in hypoparathyroidism treatment, showing improved blood calcium homeostasis compared with Matrigel-embedded dTMSCs. Interestingly, undifferentiated control TMSCs (cTMSCs) also released PTH in a sustained manner if incorporated into GHH. Collectively, these findings may establish a new paradigm for parathyroid regeneration that could ultimately evolve into an improved clinical application. Copyright (c) 2017 John Wiley & Sons, Ltd.

Characteristics of mouse embryonic fibroblasts by cryopreservation period for tissue engineering

Optimization of skeletal muscle-derived fibroblast isolation and purification without the preplating method

Summary: This study optimized the isolation and purification methods for skeletal muscle-derived fibroblasts. The results showed that the characteristics of these fibroblasts were maintained up to passage 5 regardless of preplating, and even after cryopreservation, fibroblast-specific properties remained unchanged.

Valproic Acid-Induced CCN1 Promotes Osteogenic Differentiation by Increasing CCN1 Protein Stability through HDAC1 Inhibition in Tonsil-Derived Mesenchymal Stem Cells

Summary: Our study found that the level of CCN1 increases as osteogenic differentiation progresses in tonsil-derived mesenchymal stem cells (TMSCs). We investigated how CCN1 is regulated through HDAC inhibition in TMSCs and their relationship with osteogenesis. Valproic acid (VPA), a well-known HDAC inhibitor, was found to inhibit TMSC proliferation and increase osteogenic differentiation, while decreasing adipogenesis and chondrogenesis. The effects of VPA on CCN1 were mediated through the stability of the CCN1 protein instead of gene expression. These findings demonstrate the potential of VPA as an inhibitor of HDAC1 to promote osteogenic differentiation in TMSCs.

Cyclin-Dependent Kinase 1 Inhibition Potentiates the Proliferation of Tonsil-Derived Mesenchymal Stem Cells by Delaying Cellular Senescence

Summary: Mesenchymal stem cells (MSCs) are widely used in tissue regeneration and stem cell therapy, and research on senescence-related changes in MSC properties has attracted attention. A study established a senescent MSC model and identified novel biomarkers for old, senescent tonsil-derived MSCs (TMSCs) using transcriptomics. The expression of CDK1, CDK2, and TGFB2 was significantly higher in old TMSCs compared to young TMSCs, suggesting CDK1 could be a selective senescence biomarker of MSCs to broaden the research criteria for senescent mechanisms.

Oleamide-PDMS copolymer for coating urinary catheters with anti-adhesive and anti-biofilm properties

Summary: An oleamide-polydimethylsiloxane (OPC) copolymer was synthesized to prevent biofilm infection on biomedical devices. The OPC showed anti-adhesion and anti-biofilm activities against Escherichia coli and Staphylococcus aureus. It was also found to have high biocompatibility and low hemolytic properties, making it suitable for medical device applications.

Summary: The study investigated the pathogenic mechanism leading to fibrosis in kidneys of Fabry disease mice with unilateral ureteral obstruction (UUO). The results showed an increased presence of lamellar inclusion bodies and fibrosis in the kidneys of diseased mice. Impaired autophagy and enhanced apoptosis were identified as probable mechanisms involved in the enhanced renal fibrosis observed under the stimulus of UUO in Fabry disease.
