The review highlights the use of advanced technologies such as next-generation sequencing, proteomic approaches, advanced imaging techniques, and microfluidics assays to gain insights into the underlying causes of platelet molecular, structural, and functional diversity. It discusses various sources of diversity, including different allele-specific expression patterns in proliferating megakaryocytes, asymmetrical formation of proplatelets, changes in platelets induced by aging and priming, interplatelet heterogeneity in thrombus organization and stability, and platelet-dependent communications. The review also emphasizes the importance of these technologies in characterizing platelet populations and identifying platelet biomarkers for cardiovascular disease treatment.

Summary: A combined in silico and in vitro platform is created to analyze flow conditions and cellular behavior in the onset of a hemostatic response to a microneedle-induced vessel puncture. The platform uses a cell-resolved blood flow model for detailed flow profile and cell distribution analyses, and a novel punctured vessel flow chamber to evaluate platelet aggregation around the wound neck. The simulations and experiments provide insight into platelet transport and aggregation processes in the wound area.

Summary: Platelets are small, anucleate cells in the blood that have a significant impact on both hemostasis and cardiovascular disease. Polyunsaturated fatty acids (PUFAs) are known to play a crucial role in platelet function and regulation. The oxygenase enzymes COX-1, 5-LOX, 12-LOX, and 15-LOX metabolize PUFAs into oxidized lipids (oxylipins) with either pro-thrombotic or anti-thrombotic effects. This review summarizes the known oxylipins, their activity in platelets, and current therapeutics targeting oxylipin signaling.

Engineered SERPINs hold potential as novel therapies for the treatment of thrombotic pathologies.

Summary: While cardiovascular disease (CVD) is the leading cause of death, major depressive disorder (MDD) is the primary cause of disability, affecting more than 300 million people worldwide. There is evidence that CVD is more prevalent in people with MDD. Antidepressant duloxetine, a serotonin-norepinephrine antagonist, shows potential antiplatelet and thromboprotective effects, and inhibits hemostasis, suggesting potential benefits in treating CVD associated with MDD.
