Summary: In this study, we report the first examples of thiol-selective heterobifunctional electrophiles, N-vinyl acrylamides, which enable efficient and highly selective thiol-thiol bioconjugations and cysteine modification. We demonstrate that these new thiol-selective scaffolds can undergo chemical reactions under biocompatible conditions. Computational studies explain an unexpected addition reaction. The study also reveals that N-methylation of the N-vinyl acrylamide scaffold can change the regioselectivity of the reaction.

Summary: This study reports the discovery of a highly enantioselective vinyl carbocation carbon-hydrogen (C-H) insertion reaction enabled by imidodiphosphorimidate organocatalysts. The active site confinement in this catalyst class not only allows effective enantiocontrol but also expands the scope of vinyl cation C-H insertion chemistry, enhancing the utility of this transition metal-free C(sp3)-H functionalization platform.

Summary: Energy security, job development, and minimal environmental damage are the main reasons for the popularity of green energy technologies. This study focuses on the bottlenecks in developing these technologies in underdeveloped countries, using South Africa as a case study. By utilizing a Multi-Objective Optimization based on Ratio Analysis (MOORA) strategy combined with an Evaluation based on Distance from Average Solution (EDAS) method, the study identifies social, institutional, and regulatory bottlenecks as the most impactful. Corrupt political systems, lack of public knowledge, and fixed investments are found to be the major obstacles. The study highlights the importance of a bottom-up policy approach and the need to strengthen government agencies to promote green energy technologies.

Summary: The catalytic generation of a vinyl Pd-oxyallyl that dimerizes to form highly functionalized nonbridged cyclooctanoids is reported. This compound shows both electrophilic and nucleophilic properties, and the mechanism and origins of chemoselective cyclooctanoid formation are elucidated through DFT calculations.

Summary: The C-H insertion reactions of vinyl carbocations formed from vinyl trifluoromethanesulfonates by catalytic lithiated 1,3-bis[3,5-bis(trifluoromethyl)phenyl]urea have been studied. The ion-pairing between Li+-ureide-triflate and the vinyl cation affects the thermodynamic stabilities of alkyl cations. The rate-determining step is the C-H insertion of the vinyl cation intermediate.

Summary: Metal-free cascade reaction of NH-1,2,3-triazoles with one-carbon electrophiles, such as thiophosgene and triphosgene, generates N-vinylated ring cleavage products. N-vinylisothiocyanates can be synthesized from NH-triazoles and thiophosgene using this approach. A variety of multifunctional compounds, such as N-vinylcarbamates, unsymmetrical vinylureas, carbamothioates, etc. were prepared by a one-pot method from NH-triazoles, triphosgene, and nucleophiles.

Summary: To adapt to unexpected challenges, organizations need to build and maintain the capability. However, many organizations develop slowly and can only make incremental changes. What stochastic processes govern routinely occurring challenges that help a sluggish organization prepare for unexpected challenges? We use a stylized principal-agent model to address this question and find that stochastic, time-varying challenges significantly improve a sluggish organization's readiness for sudden challenges.

Summary: Site-selective disulfide rebridging is a powerful strategy for modulating protein structure and function. We introduce oxSTEF, a novel class of electrophilic reagents, which efficiently rebridge disulfide bonds via double thiol exchange. The oxSTEF reagents show highly selective rebridging of cyclic peptides and native proteins, as well as no cross-reactivity with other nucleophilic amino acid residues. Additionally, the oxSTEF conjugates disintegrate under tumor-relevant glutathione concentrations and its α-dicarbonyl motif enables second phase oxime ligation, significantly increasing the thiol stability of the conjugates.
